富含总三萜的灵芝提取物抑制小鼠MC38结肠癌皮下移植瘤增殖的研究OA
Study on inhibition of proliferation of subcutaneously transplanted MC38 colon cancer in mice by Ganoderma lucidum extract rich in total triterpenes
目的:探讨富含总三萜的灵芝提取物抑制小鼠MC38结肠癌皮下移植瘤的作用,并分析其对髓源性抑制细胞(MDSCs)和T细胞变化的影响.方法:首先通过SwissTargetPrediction和TCMSP等数据库预测已报道文献的灵芝潜在作用靶点,并结合GeneCards数据库检索出的结肠癌相关疾病靶点推测灵芝作用于结肠癌的潜在靶点;运用STRING数据库构建交集靶点的蛋白互作(PPI)网络;使用Cytoscape软件对STRING数据库筛选的靶点进行网络拓扑分析筛选关键靶点,并推测潜在药物活性成分;运用DAVID数据库对差异基因进行GO和KEGG通路富集分析.基于上述推测的灵芝抑制肿瘤的潜在重要活性成分结果,建立MC38细胞系皮下结肠癌小鼠模型,灌胃不同总三萜含量的灵芝提取物.Ki67免疫组化染色观察小鼠肿瘤细胞增殖能力;流式细胞术检测小鼠肿瘤及脾脏中T细胞及MDSCs变化;ELISA检测小鼠血清中IL-6表达.结果:通过网络药理学分析已报道的灵芝提取物抑制结肠癌的活性成分,发现灵芝提取物中抑制结肠癌的主要活性成分属于总三萜类.分别灌胃荷瘤小鼠N(生理盐水)、制剂A(总三萜0.01%)、制剂B(总三萜0.2%)和制剂C(总三萜3.5%),结果表明,与N组相比,制剂C灌胃后的结肠癌小鼠肿瘤生长明显变慢,肿瘤体积明显减小;ELISA结果表明,制剂C可降低小鼠血清IL-6水平;流式细胞术结果显示C组小鼠肿瘤和脾脏中G-MDSCs比例降低,CD8+T细胞比例增加.结论:富含总三萜的灵芝提取物可明显抑制MC38细胞系皮下结肠癌小鼠模型肿瘤增殖,显著下调荷瘤小鼠瘤内和脾脏中G-MDSCs比例,提高CD8+T细胞比例,提示富含总三萜的灵芝提取物抑制结肠癌的作用主要是通过调节免疫功能实现的.
Objective:To investigate effects of Ganoderma lucidum extract rich in total triterpenes on inhibition of subcutane-ous colon cancer in mice with MC38 cell line,and analyze its impact on changes in myeloid-derived suppressor cells(MDSCs)and T cells.Methods:Initially,potential targets of Ganoderma lucidum reported in literature were predicted using databases such as Swiss TargetPrediction and TCMSP,and potential targets of colon cancer were inferred by combining disease targets related to colon cancer retrieved from GeneCards database;STRING database was utilized to construct a protein-protein interaction(PPI)network for inter-secting targets;Cytoscape software was employed for network topology analysis of targets selected from STRING database to identify key targets and speculate on potential active drug components;DAVID database was used to perform GO and KEGG pathway enrich-ment analysis on differential genes.Based on results of inferred potential important active components of Ganoderma lucidum in inhibiting tumor growth,a subcutaneous colon cancer mouse model with MC38 cell line was established and administered intragastric treatment with Ganoderma lucidum extracts of varying total triterpene content.Ki67 immunohistochemical staining was used to observe prolifera-tive capacity of mouse tumors;flow cytometry was used to determine changes in T cells and MDSCs in tumors and spleens of mice;ELISA was used to measure level of IL-6 in mouse serum.Results:Network pharmacology analysis revealed that main active compo-nents in Ganoderma lucidum extract that inhibit colon cancer belong to class of total triterpenes.Mice bearing tumors were adminis-tered intragastric treatments with N(normal saline),Formulation A(0.01%total triterpenes),Formulation B(0.2%total triterpenes)and Formulation C(3.5%total triterpenes).Compared to N group,growth of tumors in colon cancer mice treated with Formulation C was significantly slowed,and tumor volume was markedly reduced.ELISA results demonstrated that Formulation C could decrease expression of IL-6 in mouse serum.Flow cytometry results showed that proportion of G-MDSCs in tumors and spleens of mice in Group C was decreased,while proportion of CD8+T cells was increased.Conclusion:Ganoderma lucidum extract rich in total triterpenes can significantly inhibit growth of colon cancer in MC38 cell line subcutaneous mouse model,markedly down-regulate proportion of G-MDSCs in tumors and spleens of tumor-bearing mice,and increase proportion of CD8+T cells,suggesting that inhibitory effect of Ganoderma lucidum extract rich in total triterpenes on colon cancer is mainly achieved through modulation of immune responses.
