不同杂交组合黄羽肉鸡皮肤毛囊密度差异及其与Wnt3a表达水平的相关性分析OA
Skin feather follicle density and Wnt3a mRNA expression in different hybrid combinations of yellow-feathered chicken
为探究黄羽肉鸡皮肤毛囊密度性状的遗传差异及其分子机制,本研究以来源于地方鸡种和快大型黄鸡配套系的4个专门化品系(K系、T系、W系和R系)为素材,按不完全双列杂交法设计形成6个二元杂交组合(WT、RT、KW、TW、KR和TR),测定不同杂交组合公、母鸡300日龄背部、胸部和腿部皮肤毛囊密度和Wnt3a mRNA表达量,并进行相关性分析.结果表明:1)不同杂交组合之间体重存在差异,但差异不显著(P>0.05),以来源于快大型黄鸡(W、R系)为父本的组合体重高于其他组合;2)不同杂交组合之间皮肤毛囊密度存在显著差异(P<0.05),以来源于地方鸡种(K、T系)为父本的组合毛囊密度高于其他组合;3)各组合不同部位皮肤毛囊密度和Wnt3a mRNA表达量存在显著差异(P<0.05),均呈背部>胸部>腿部的梯度分布规律,且二者呈不同程度正相关;4)各组合皮肤毛囊密度与体重大小均呈不同程度负相关.综上,本研究通过比较不同杂交组合黄羽肉鸡皮肤毛囊密度差异,验证了 Wnt3a对皮肤毛囊发育的关键调控作用,揭示了生长速度与毛囊发育之间可能存在的遗传拮抗关系,为优质鸡选育过程中平衡生长速度与胴体外观性状的分子育种策略提供了理论依据.
The present study was conducted with the objective of investigating the differences in skin feather follicle density and its correlation with Wnt3a mRNA expression in different hybrid combinations of yellow-feathered chickens.The experiment was conducted on the basis of four specialized strains,which were bred from local native chicken breeds and fast-sized yellow chicken resources.A total of six binary hybrid combinations(WT,RT,KW,TW,KR and TR)were designed by incomplete double-row crosses.The skin follicle density and Wnt3a mRNA expression levels of diverse skin regions(i.e.back,breast and thigh)of these hybrid combinations were systematically assessed,and a comprehensive correlation analysis was conducted.The results demonstrated that:1)The body weights of the different hybrids were not significantly different(P>0.05),and the body weights of the hybrids sired by fast-sized yellow chicken(W and R strains)were higher than the others.2)The feather follicle densities of the various hybrids exhibited significant variation(P<0.05),with the follicle density of the hybrids sired by the local breeds(K and T strains)being higher than that of the other hybrids.3)A statistically significant discrepancy(P<0.05)was identified in the feather follicle density and Wnt3a mRNA expression levels among the distinct anatomical regions of the hybrid subjects.A gradient distribution pattern was observed,with higher levels in the back region followed by the breast and then the thigh.A positive correlation was identified between follicle density and Wnt3a mRNA expression,with varying degrees across different hybrids.4)A negative correlation was identified between feather follicle density and body weight with varying degrees across different hybrids.In summary,the present study compared the differences in skin feather follicle density among different hybrid combinations of yellow-feathered chickens,confirming the regulatory role of the Wnt3a gene in feather follicle development and revealing the potential existence of a genetic antagonism between growth rate and feather follicle development,which provided a theoretical basis for the molecular breeding strategy to balance growth rate and carcass appearance traits in the selection and breeding of high-quality yellow-feathered chickens.
