新藤黄酸对喉癌细胞增殖、迁移的抑制作用及其机制研究OA
Inhibitory Effects of Gambogenic Acid on Proliferation and Migration of Laryngeal Cancer Cells and Its Mechanism
目的 探究天然化合物新藤黄酸对喉癌(Tu686)细胞增殖、迁移的影响及其作用机制.方法 采用MTT法、平板克隆、划痕实验、Transwell实验、Hoechst 33258 染色等方法,分析新藤黄酸对Tu686 细胞生长、迁移的影响.网络药理学及分子对接预测新藤黄酸作用于喉癌细胞的分子靶点,最后Western blot实验检测相关蛋白的表达水平并予以验证.结果 新藤黄酸可显著抑制Tu686 细胞的增殖以及迁移、侵袭能力.网络药理学筛选获得新藤黄酸-喉癌共同靶点 19个,KEGG富集分析提示,新藤黄酸可能通过调控癌症相关通路等抑制肿瘤进程.分子对接结果显示,新藤黄酸与核心靶点具有强结合活性.Hoechst 33258 染色和Western blot结果表明,新藤黄酸可以诱导Tu686 细胞凋亡,并且还可使STAT3、BCL2 蛋白表达下调,BAX蛋白表达上调.结论 新藤黄酸对Tu686 细胞的增殖、迁移能力有明显的抑制作用,并且可能与多种靶点有关.
Objective To investigate the effect of gambogenic acid on the proliferation and migration of laryngeal cancer Tu686 cells and its mechanism.Methods The effects of gambogenic acid on the growth and migration of Tu686 cells were analyzed by MTT assay,plate cloning assay,wound healing assay,Transwell assay and Hoechst 33258 staining.The molecular targets of gambogenic acid on laryngeal cancer cells was predicted by Network pharmacology and molecular docking,and the expression levels of related proteins were detected by Western blot.Results Gambogenic acid significantly inhibited the proliferation,migration and invasion of Tu686 cells.Network pharmacology screening identified 19 common targets of gambogenic acid-laryngeal cancer.KEGG enrichment analysis suggested that gambogenic acid might inhibit tumor progression by regulating cancer-related pathways.Molecular docking results showed that gambogenic acid had strong binding activity to the core targets.Hoechst 33258 staining and Western blot results showed that gambogenic acid could induce apoptosis of Tu686,down-regulate the expression of STAT3 and BCL2 proteins and up-regulate the expression of BAX protein.Conclusion Gambogenic acid can significantly inhibit the proliferation and migration of laryngeal cancer Tu686 cells,which may be related to multiple targets.
