为开发能够调节动物氧化应激、缓解肠道炎症的益生菌制剂,通过体外抗氧化试验,从野猪新鲜粪便中分离筛选到 1 株植物乳杆菌(Lac⁃tobacillus plantarum),菌株编号为9-4.采用牛津杯扩散法检测该菌株的抑菌能力,并评估其对人工胃肠液和胆盐的耐受能力;动物试验以 3%葡聚糖硫酸钠(DSS)构建小鼠结肠炎模型,分析L.plantarum 9-4 干预对结肠炎小鼠临床症状、血清氧化应激水平和肠道炎症的影响.结果:L.plantarum 9-4 对大肠杆菌、沙门菌和金黄色葡萄球菌均有抑制作用,并具有良好的耐酸、耐胆盐特性.动物试验表明,L.plantarum 9-4 可极显著提高结肠炎小鼠体重(P<0.001),极显著降低疾病活动指数(DAI)(P<0.001),并有效抑制结肠长度缩短(P<0.05);干预后小鼠血清丙二醛(MDA)含量极显著降低(P<0.01),超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)水平有升高趋势;结肠组织中肿瘤坏死因子α(TNF-α)、白细胞介素 1β(IL-1β)和白细胞介素 6(IL-6)的mRNA表达量均显著下降(P<0.05).综上,L.plantarum 9-4 能发挥抗氧化和抗炎效果,具有开发为益生菌制剂用于畜禽养殖的潜力.

To develop probiotic products capable of regulating oxidative stress and alleviating intestinal inflammation in animals,Lactobacil⁃lus plantarum strain 9-4 was isolated and screened from fresh wild boar feces through in vitro antioxidant tests.Antibacterial activity of the strain was detected via the Oxford cup method,and its tolerance to artificial gastrointestinal fluids and bile salts was evaluated.Then,a mouse colitis model was established using 3%dextran sulfate sodium(DSS),and the effects of L.plantarum 9-4 intervention on clinical symptoms,intestinal inflammation,and serum oxidative stress levels in colitis mice were analyzed.The results were as follows:L.plantarum 9-4 exhibited inhibitory effects on Escherichia coli,Salmonella,and Staphylococcus aureus,and maintained a high survival rate in artificial gastric juice,artificial intestinal fluid,and 0.3%bile salt.Intervention with L.plantarum 9-4 significantly increased the body weight of the colitis mice(P<0.001),reduced the disease activity index(DAI)(P<0.001)in the rodents,and attenuated their colon length shortening(P<0.05).Additionally,L.plantarum 9-4 intervention significantly decreased the serum malondialdehyde(MDA)levels in the mice(P<0.01),while the superoxide dismutase(SOD),catalase(CAT),and glutathione peroxidase(GSH-Px)levels in them showed an upward trend.Moreover,the mRNA expression levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6)in the colon tissue of the mice were significantly downregulated(P<0.05).In conclusion,L.plantarum 9-4 possessed antioxidant and anti- inflammatory effects and promised a potential to be developed into probiotic preparation for use in livestock and poultry farming.
