基于HaCaT和RAW264.7细胞模型的D-泛醇修护和舒缓功效机制研究OA
Mechanistic study of the repairing and soothing effects of D-panthenol using HaCaT and RAW264.7 cell models
分别以H2O2刺激HaCaT细胞和LPS刺激RAW264.7细胞为模型,探究D-泛醇的屏障修护机制和抗炎机制.结果表明,0.08%~0.31%和0.08%~1.25%浓度范围内的D-泛醇分别对HaCaT和RAW264.7细胞均无毒性;同时,0.31%D-泛醇能够显著提升H2O2刺激下HaCaT细胞中FLG和TGM1基因的表达,提升率分别为114.79%和66.41%;1.25%D-泛醇对LPS刺激的RAW264.7细胞释放NO、PGE2、TNF-ɑ、IL-1β和IL-6存在显著抑制作用,抑制率分别为28.33%、41.75%、32.82%、75.93%和43.83%.D-泛醇在安全作用浓度下对屏障基因表达的提升作用和对炎症因子的抑制作用提示其具有潜在的修护和舒缓皮肤的功效,进一步为D-泛醇在皮肤护理领域的应用提供理论依据.
Barrier-repairing and anti-inflammatory mechanisms of D-panthenol using H2O2-stimulated HaCaT cells and LPS-stimulated RAW264.7 cells as in vitro models were investigated.D-panthenol at concentrations of 0.08%~0.31%and 0.08%~1.25%exhibited no cytotoxicity to HaCaT and RAW264.7 cells,respectively.Notably,treatment with 0.31%D-panthenol significantly increased the expression of FLG and TGM1 genes in H2O2-stimulated HaCaT cells by 114.79%and 66.41%,respectively.Additionally,1.25%D-panthenol markedly inhibited the release of NO,PGE2,TNF-α,IL-1β,and IL-6 in LPS-stimulated RAW264.7 cells,with inhibition rates of 28.33%,41.75%,32.82%,75.93%,and 43.83%,respectively.These findings indicate that D-panthenol enhances barrier gene expression and suppresses inflammatory mediator production at safe concentrations,supporting its potential use in skin repair and soothing applications.This study provides further theoretical evidence for the incorporation of D-panthenol in skincare formulations.
