6种根际促生细菌对浙贝母生长与活性成分的影响OA

[目的]本文旨在从浙贝母(Fritillaria thunbergii)根际土壤中分离促生菌,研究 6 种不同根际促生菌对浙贝母生长及活性成分的影响,为浙贝母化肥、农药的减量施用和生态农业种植提供依据.[方法]选择各类特异培养基筛选根际土壤细菌的固氮、溶磷、解钾、产IAA和产铁载体能力;采用6 种浙贝母促生菌(沙雷氏菌TZ1、芽胞杆菌JJ2、微小杆菌JJ15、大肠杆菌TZ21、泛菌TZ112 和假单胞菌JJ137)单施的方法开展盆栽试验,测定浙贝母生长指标、叶绿素、渗透调节物质、活性成分等水平,探究根际促生菌对浙贝母的促生效果.[结果]从浙贝母根际土壤中筛选到 6 种根际促生细菌,6 种促生菌对不同生长时期浙贝母生长均有显著影响.6 种根际细菌均可促进浙贝母生长,增加叶、茎、鳞茎的干重、鲜重等,始枯期TZ1 组的叶面积较CK组(不接种根际菌)增加 28.69%,始枯期TZ112 组的株高较CK组提高 23.75%.根际促生菌对浙贝母叶片叶绿素和渗透调节物质含量也有显著影响,始枯期TZ1 组的叶绿素a、叶绿素b、总叶绿素含量增加均最为显著,较CK组分别增加 14.70%、15.69%、14.51%.6 种根际细菌均有利于休眠期浙贝母活性成分(贝母素甲、贝母素乙)的积累,其中TZ112组单株鳞茎生物量、贝母素乙含量、单株总活性成分生物量增加均最为显著,较CK组分别增加 18.83%、30.12%、33.01%.[结论]浙贝母根际存在具有多种促生特性的细菌,分离筛选的菌株TZ1、JJ2、JJ15、TZ21、TZ112 和JJ137 均具促生能力,对浙贝母的生长与活性成分的增加有促进作用,其中TZ112 的综合效果最佳.

[Objectives]The paper aimed to isolate the growth-promoting bacteria from the rhizosphere soil of Fritillaria thunbergii.The effects of 6 different rhizosphere growth-promoting bacteria on growth and active components of F.thunbergii were studied in order to provide basis for fertilizer and pesticide reduction application and ecological agriculture planting.[Methods]Various specific media were selected to screen the capacities of nitrogen fixation,phosphorus solubilization,potassium solubilization,IAA production and iron production of rhizosphere growth-promoting bacteria.Six growth-promoting bacteria(Serratia sp.TZ1,Bacillus sp.JJ2,Exiguobacterium sp.JJ15,Enterobacter sp.TZ21,Pantoea sp.TZ112 and Pseudomonas sp.JJ137)were applied in pot experiment,and the growth indexes,chlorophyll,osmotic regulatory substances and active components were determined to explore the growth promotion effect of rhizosphere growth-promoting bacteria on F.thunbergii.[Results]Six rhizosphere growth-promoting bacteria were screened from rhizosphere soil of F.thunbergii.Six growth-promoting bacteria had all significant effects on growth of F.thunbergii at different growth stages.Six rhizosphere bacteria could promote the growth of plant and increase the dry weight and fresh weight of leaves,stems,bulbs,etc.The leaf area of TZ1 group was 28.69%higher than CK group(no inoculation with soil rhizobacteria)at the beginning of depletion stage,and plant height of TZ112 group were 23.75%higher than CK group at the beginning of depletion stage.Rhizosphere growth-promoting bacteria also had significant effects on the contents of chlorophyll and osmoregulatory substances in the leaves of F.thunbergii.The contents of chlorophyll a,chlorophyll b and total chlorophyll in TZ1 group at the beginning of depletion stage increased most significantly,which were 14.70%,15.69%and 14.51%higher than those in CK group,respectively.Six rhizosphere bacteria were beneficial to the accumulation of active components(peimine and peiminine)of F.thunbergii during the dormant period.Among them,bulb biomass per plant,peiminine content and total active component biomass per plant in TZ112 group increased most significantly,which were 18.83%,30.12%and 33.01%higher than those in CK group,respectively.[Conclusions]There were growth-promoting bacteria with a variety of characteristics in the rhizosphere of F.thunbergii.The isolated and screened bacteria TZ1,JJ2,JJ15,TZ21,TZ112 and JJ137 had the ability to promote the growth and physiological process of F.thunbergii,among which TZ112 had the best comprehensive capability.
