[目的]本文旨在探讨胎生睡莲株龄对叶片生长和叶脐胎芽退化的影响.[方法]以1~4 年生'黑美人'睡莲叶片为材料,测定不同株龄的叶片形态参数,结合石蜡切片法观察相应时期的叶片、叶脐和叶脉组织结构变化,并比较叶片和叶脐部位碳水化合物及蔗糖代谢酶活性的差异.[结果]随着株龄的增加,'黑美人'叶面积和叶片厚度不断增加,叶绿素和类胡萝卜素含量均先升后降,而胎芽衰退的叶片叶绿素含量下降减缓;相较于不同年龄的叶片解剖结构,1~4 年生胎生叶脐部位凸起且细胞排列紧密,而胎芽消失的 4 年生叶脐表面光滑,表皮细胞排列较疏松,但叶片和叶脉组织在结构上无明显差异.'黑美人'叶片和叶脐的可溶性蛋白含量及过氧化物酶、过氧化氢酶和蔗糖合成酶活性随株龄的增加呈先升后降的趋势,而可溶性糖和还原糖含量则先降低后升高,淀粉含量及超氧化物歧化酶和蔗糖磷酸合成酶活性则逐渐降低,这些指标均表现为叶片大于叶脐,但胎芽退化后的叶脐与正常的叶脐差异不大.[结论]睡莲胎生叶片(源)与叶脐(库)的关系随年龄发生动态变化,且胎芽的退化明显受株龄的影响,叶片不再具有胎生繁殖能力,但整个植株尚未进入衰老状态.因此,胎芽的退化可能受到精细控制局部细胞死亡事件的调控.

[Objectives]This study aimed to explore the effects of different plant ages of tropical viviparous water lily on leaf growth and epiphyllous bud degeneration.[Methods]The leaves of one to four-year-old potted'Black Beauty'as materials,the morphological parameters of leaves were measured at different ages,combined with paraffin sectioning,the tissue structure changes of leaves,leaf navels,and leaf veins at corresponding periods were observed,and the differences in carbohydrate and sucrose metabolic enzyme activities between the leaves and leaf navels were compared.[Results]As the plant age increased,the leaf area and thickness of'Black Beauty'continuously increased,and the chlorophyll and carotenoid contents first increased and then decreased.However,the chlorophyll content in leaves with epiphyllous bud degeneration decreased.Compared with the anatomical structures of leaves at different ages,the leaf navels of one to four years old were raised and the cells were tightly arranged,while the surface of the four-year-old leaf navel of epiphyllous bud decline was smooth with relatively loose epidermal cells.However,there was no significant difference in the structure of leaves and leaf veins.The soluble protein content and the activities of peroxidase,catalase and sucrose synthase in the leaf and leaf navel of'Black Beauty'increased first and then decreased with age.Meanwhile,the contents of soluble sugar and reducing sugar initially decreased and then increased,while the starch content,superoxide dismutase activity and sucrose phosphate synthase activity consistently decreased.These indicators showed that the leaves were greater than the leaf navels,but there was no significant difference between the degenerated epiphyllous bud and the normal one.[Conclusions]The relationship between viviparous leaves(source)and leaf navel(sink)of water lilies varied with age and was influenced by plant age.Leaves no longer had the viviparous ability,but the entire plant had not entered a senescence state.Therefore,the epiphyllous bud degeneration might be regulated by finely controlling local cell death events.
