基于块染技术的电镜生物样本免切片染色制备方法探究OA
Exploration of sectioning-free staining preparation methods for biological samples in electron microscopy based on en-block staining techniques
合适的块染制样方法是生物体电子显微成像质量的关键因素.本研究以小鼠肾脏为模型,在常规生物样本块染基础之上,系统研究了 13种直接利用透射电镜(TEM)观察的生物样本包埋前进行块染制备方法.结果显示,与常规铀块染方法相比,不同染色试剂及染色顺序会影响超微结构观察.其中,四氧化锇-硫代甲酰二肼-四氧化锇(OTO法)、四氧化锇-单宁酸-单宁酸-四氧化锇(OTTO法)及亚铁氰化钾-四氧化锇-硫代甲酰二肼-四氧化锇(ROTO法)所制得的样本,提高了线粒体等细胞器的结构清晰度及图像反差;重金属铅、铀的染色顺序也会影响样本线粒体嵴结构清晰度;优化后的亚铁氰化钾-四氧化锇-单宁酸-单宁酸-四氧化锇[ROT(T)O法]结合铅、铀双重块染,不仅能增加样品衬度,同时更有效地保持细胞骨架和细胞质膜结构,其成像效果最佳.
The appropriate block staining method is crucial for ensuring the quality of biological electron microscopy imaging.Based on conventional biological sample block staining,this study systematically investigated 13 direct block staining methods for biological samples that can be observed with a transmission electron microscope(TEM)before embedding,using mouse kidneys as a model.The results showed that the observation of ultrastructure was affected by different staining reagents and staining sequences,compared with the conventional uranium block staining method.Among them,the samples prepared by the osmium tetroxide-thiocarbohydrazide-osmium tetroxide(OTO method),the osmium tetroxide-tannic acid-tannic acid-osmium tetroxide(OTTO method),and the potassium ferrocyanide-osmium tetroxide-thiocarbohydrazide-osmium tetroxide(ROTO method)showed improved structural clarity and image contrast of organelles such as mitochondria.The staining sequence of heavy metals lead and uranium was found to affect the clarity of mitochondrial cristae structure.The optimized potassium ferrocyanide-osmium tetroxide-tannic acid-tannic acid-osmium tetroxide(ROT(T)O method),when combined with lead and uranium double block staining,not only increased the sample contrast,but also more effectively maintained the cytoskeleton and cell membrane structure,thereby achieving the best imaging effect.
