基于ISSR分子标记的云锦杜鹃遗传多样性研究OA
Genetic diversity of Rhododendron fortunei based on ISSR molecular marker
采用ISSR分子标记技术对我国4个云锦杜鹃自然居群的遗传多样性进行了研究.实验结果表明:8条引物共扩增出189个多态位点,多态性位点比率为98.44%;物种水平的Nei's基因多样性指数、Shannon's信息指数、观测等位基因数和有效等位基因数分别为0.233 1、0.374 5、1.989 6和1.364 0;居群水平的遗传多样性指数为0.235 0,居群间的平均遗传多样性指数为0.068 1,居群内的平均遗传多样性指数为0.166 9,遗传分化系数为0.290 0,这些指数表明云锦杜鹃的遗传多样性水平较低;AMOVA分子差异分析显示29%的变异存在于居群间,71%的变异存在于居群内,表明云锦杜鹃居群内产生了显著的遗传变异,居群间的遗传分化较小;云锦杜鹃居群间的基因流为1.224 4>1,表明居群间存在一定的基因流,可以防止遗传漂变引起的遗传分化;基于遗传距离的4个云锦杜鹃居群聚类分析表明,广西十里平坦居群与江西井冈山居群的亲缘关系较近,两居群间基因交流程度高;广西猫儿山居群与其他三个居群的亲缘关系较远,遗传分化程度较高.
The genetic diversity of four natural populations of Rhododendron fortunei in China was studied using the inter-simple sequence repeat(ISSR)molecular marker technique.The results showed that a total of 189 polymorphic sites were amplified by 8 primers,and the polymorphic site ratio was 98.44%.The Nei's gene diversity index,Shannon's information index,the number of observed alleles and effective alleles at the species level were 0.233 1,0.374 5 1.989 6 and 1.364 0,respectively.The genetic diversity index at the population level was 0.235 0,the average genetic diversity index among populations was 0.0681,the average genetic diversity index within the population was 0.166 9,and the coefficient of genetic differentiation was 0.290 0,which indicated that the genetic diversity level of R.fortunei was low.The AMOVA molecular difference analysis showed that 29%of the variation existed between populations,and 71 of the variation existed within populations,that significant genetic variation had occurred within the populations of R.fortunei,and the genetic differentiation among populations was small;the gene flow among the populations of R.fortunei was 1.224 4>1,indicating that was a certain degree of gene flow among the populations,which could prevent the genetic differentiation caused by genetic drift.The cluster analysis based on genetic distance of the four populations of R.fortunei showed that the population in Guangxi Shilipingtan was closely related to the population in Jiangxi Jinggangshan,and the gene exchange level between the two populations was high;the population in Guangxi Maoershan was distantly related to the other three populations,and the degree of genetic differentiation was high.
