和胃安神方对失眠模型大鼠视交叉上核c-fos、c-jun及神经元型一氧化氮合酶表达的影响OA
Effects of Hewei Anshen Formula on the expression of c-fos,c-jun,and nNOS in the suprachiasmatic nucleus of insomnia model rats
目的 探讨和胃安神方对失眠模型大鼠视交叉上核(suprachiasmatic nucleus,SCN)即刻早期基因c-fos、c-jun以及神经元型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)表达的影响.方法 选用 60 只雄性SD大鼠,随机分配至正常对照组、模型对照组、艾司唑仑组以及和胃安神方低、中、高剂量组,每组10 只.除正常对照组外,其余各组大鼠均通过腹腔注射对氯苯丙氨酸诱导失眠模型.造模效果通过戊巴比妥钠翻正实验进行评估.造模成功后,各组大鼠接受连续7 天的灌胃给药:正常对照组与模型对照组灌服10 mL/kg蒸馏水;艾司唑仑组给予0.33 mg/(kg·d)艾司唑仑水溶液;和胃安神方低、中、高剂量组则分别灌胃浓度为 1 g/mL、2 g/mL、4 g/mL的和胃安神方药液,给药体积均为10 mL/kg.给药7 天完成后,开展旷场实验及高架十字迷宫实验以评估大鼠行为并采用免疫组化法和qPCR法检测大鼠SCN中c-fos、c-jun以及nNOS表达情况.结果 造模结束后,与正常对照组相比,造模组大鼠睡眠潜伏期延长(P<0.01),睡眠时间缩短(P<0.01).末次给药后,与正常对照组相比,模型对照组大鼠旷场运动总路程、平均速度显著增加(P<0.01),旷场中央区停留时间显著下降(P<0.01),高架十字迷宫开放臂进入次数比例、开放臂停留时间比例下降(P<0.01),SCN中c-fos、c-jun及nNOS的表达水平显著升高(P<0.01).与模型对照组相比,艾司唑仑组与和胃安神方中、高剂量组大鼠旷场运动总路程、平均速度显著下降(P<0.05 或P<0.01),旷场中央区停留时间显著上升(P<0.05 或P<0.01);艾司唑仑组与和胃安神方低、中、高剂量组大鼠高架十字迷宫进入开放臂次数比例、开放臂停留时间比例均升高(P<0.05 或P<0.01),SCN中c-fos、c-jun及nNOS的表达水平显著降低(P<0.05 或P<0.01).结论 和胃安神方能有效缓解失眠模型大鼠的睡眠障碍,可能与其对SCN中c-fos、c-jun及nNOS表达水平的下调作用相关.
Objective To investigate the effects of Hewei Anshen Formula on the expression of im-mediate early genes(IEGs)c-fos,c-jun,and neuronal nitric oxide synthase(nNOS)in the suprachiasmatic nucleus(SCN)of insomnia model rats.Methods Sixty male Sprague-Dawley rats were randomly allocated to six groups(n=10 per group):normal control,model control,estazolam,and Hewei Anshen Formula low-,medium-,and high-dose groups.Except for the normal control group,insomnia models were induced in all other groups via intraperitoneal injection of para-chlorophenylalanine(PCPA),with modeling efficacy validated by pentobarbital sodium-induced righting reflex test.Following successful modeling,rats received 7 consecutive days of intragastric administration:the normal control and the model control groups were administered 10 mL/kg distilled water;the estazolam group received 0.33 mg/(kg·day)estazolam aqueous solution;Hewei Anshen Formula groups were given Hewei Anshen Formula decoction at concentrations of 1 g/mL(low-dose),2 g/mL(medium-dose),and 4 g/mL(high-dose),all at 10 mL/kg volume.Upon completion of treatment,behavioral assessments were conducted using open field test(OFT)and elevated plus maze(EPM)test,while expression of c-fos/c-jun proteins and c-fos/c-jun/nNOS genes in the SCN were analyzed by immunohistochemistry(IHC)and quantitative polymerase chain reaction(qPCR),respectively.Results After model establishment,compared with the normal control group,the model groups showed significantly prolonged sleep latency(P<0.01)and reduced total sleep time(P<0.01).Following the final drug administration,the model control group exhibited significantly increased total distance traveled(P<0.01)and average speed(P<0.01)in the open field test(OFT),while displaying significantly decreased central area duration(P<0.01)in OFT,reduced percentage of open arm entries(P<0.01)and open arm time(P<0.01)in the elevated plus maze(EPM),along with significantly elevated expression of c-fos/c-jun proteins and c-fos/c-jun/nNOS genes in the suprachiasmatic nucleus(SCN)(all P<0.01).Compared with the model control group,both the estazolam group and the Hewei Anshen Formula medium/high-dose groups demonstrated significantly decreased total distance(P<0.05 or P<0.01)and average speed(P<0.05 or P<0.01)in OFT with increased central area duration(P<0.05 or P<0.01),whereas the estazolam group and all the Hewei Anshen Formula dose groups showed increased percentage of open arm entries(P<0.05 or P<0.01)and open arm time(P<0.05 or P<0.01)in EPM,accompanied by significantly reduced expression of c-fos/c-jun/nNOS in SCN(P<0.05 or P<0.01).Conclusion Hewei Anshen Formula ameliorates insomnia in rats,potentially through down-regulating the expression of c-fos,c-jun,and nNOS in the SCN.
