目的 探究甘草及其不同极性部位对附子中三种生物碱(乌头碱、次乌头碱、苯甲酰新乌头原碱)在不同环境条件下的体外稳定性影响.方法 采用超高效液相色谱(ultra-high performance liquid chromatography,UPLC)分析不同条件(pH、温度)下三种生物碱的含量变化,以化学动力学方法拟合其降解规律并计算相关动力学参数.结果 三种生物碱在碱性(pH 8.3)条件下降解明显,且符合一级动力学模型.甘草提取液一定程度上提高了3 种生物碱的稳定性,对次乌头碱的促稳作用最显著.三种生物碱在高温条件下的降解(60℃、70℃和90℃)符合一级动力学模型,甘草提取液显著延缓了降解速率;甘草提取液及其不同极性部位能促进乌头碱和次乌头碱在pH 8.3 条件下的稳定性,但对苯甲酰新乌头原碱的促稳作用不显著.结论 甘草及其不同极性部位可不同程度影响附子中 3 种主要生物碱的稳定性,其中以甘草提取液最为显著,从生物碱的稳定性角度阐释了甘草与附子配伍的科学内涵.

Objective To investigate the effects of licorice and its different polar parts on the in vitro stabil-ity of three alkaloids(aconitine,hypaconitine,benzoylmesaconine)in Aconiti Radix Lateralis Praeparata under different environmental conditions.Methods Ultra performance liquid chromatography was used to analyze the content changes of three alkaloids under different conditions(pH,temperature),and the chemical kinetic method was applied to fit the degradation kinetics and calculate the relevant kinetic parameters.Results The three alkaloids degraded significantly under alkaline conditions(pH 8.3)and conformed to the first-order ki-netic model.The licorice extract improved the stability of the three alkaloids to a certain extent,and had the most significant effect on the stability of hypaconitine.The degradation of three alkaloids under high temperature conditions(60℃,70℃and 90℃)conformed to the first-order kinetics,and the licorice extract significantly delayed the degradation rate.The licorice extract and its different polar components could promote the stability of aconitine and hypaconitine at pH 8.3,but had no significant effect on benzoylmesaconine.Conclusion Lico-rice and its different polar parts can affect the stability of the three main alkaloids in Aconiti Radix Lateralis Pra-eparata to varying degrees,among which licorice extract showed the most prominent effect.The scientific conno-tation of the compatibility of licorice and Aconiti Radix Lateralis Praeparata is interpreted from the perspective of stability.
