桑麻宣肺止咳方对感染后咳嗽大鼠气道炎症和氧化应激的影响OA
Effects of Sangma Xuanfei Zhike Formula on Airway Inflammation and Oxidative Stress in Rats with Post-Infectious Cough
目的 探讨桑麻宣肺止咳方对感染后咳嗽(post-infectious cough,PIC)大鼠气道炎症的改善作用及相关机制.方法 将60 只SD雄性大鼠,随机分为正常组,模型组,桑麻宣肺止咳方(低、中、高)剂量组,对照组.除正常组外,其余组采用烟熏+鼻滴脂多糖+辣椒素雾化构建PIC大鼠模型.观察咳嗽情况,苏木精-伊红染色法观察肺组织病理变化,酶联免疫吸附分析法(enzyme-linked immunosorbent as-say,ELISA)检测血清白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)表达;试剂盒检查血清丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)浓度;蛋白质印迹法(Western blot,WB)检测肺组织核因子-κB(nuclear factor-κB,NF-κB)、胱天蛋白酶1(caspase-1)、IL-1β、白细胞介素-18(interleukin-18,IL-18)蛋白表达.结果 与正常组相比,模型组大鼠肺组织和气道炎症细胞浸润明显,血清炎症因子IL-1β、IL-6、TNF-α表达明显上升(P<0.05),肺组织MDA含量明显上升,CAT、SOD含量明显降低(P<0.05),肺组织炎症调控相关蛋白NF-κB、caspase-1、IL-1β、IL-18 表达明显上升(P<0.05);给药后,与模型组比较,桑麻宣肺止咳方可减轻PIC模型大鼠肺组织气道炎症等病理变化,降低血清IL-1β、IL-6、TNF-α、MDA和升高CAT、SOD的含量,降低肺组织NF-κB、caspase-1、IL-1β、IL-18 蛋白的表达.结论 桑麻宣肺止咳方可能通过下调NF-κB、caspase-1、IL-1β、IL-18 表达,抑制了炎症因子的分泌,减轻肺组织病理改变,从而有效改善PIC大鼠咳嗽症状.
Objective To investigate the ameliorative effect and the related mechanism of Xuanfei Zhike Formula(SMXFZKF)on airway inflammation in rats with post-infectious cough(PIC).Methods Sixty male SD rats were randomly divided into a normal group,a model group,a low-,medium-,and high-dose SMXFZ-KF groups,and a positive control group.Except for the normal group,PIC rat models were established in the other groups using smoke inhalation+intranasal lipopolysaccharide(LPS)+capsaicin aerosol exposure.Coug-hing behavior was observed.Pathological changes in lung tissues were examined by using hematoxylin-eosin(HE)staining.Serum levels of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were measured by using enzyme-linked immunosorbent assay(ELISA).Serum concentrations of ma-londialdehyde(MDA),superoxide dismutase(SOD),and catalase(CAT)were determined by using assay kits.Protein expression of nuclear factor-κB(NF-κB),caspase-1,IL-1β,and interleukin-18(IL-18)in lung tissue was detected by Western blot.Results Compared with the normal group,the model group showed significant inflammatory cell infiltration in lung tissues and airways,significantly elevated serum levels of the in-flammatory cytokines IL-1β,IL-6,and TNF-α(P<0.05),marked elevated MDA content in lung tissue,and notable reduced levels of CAT and SOD(P<0.05).The expression of inflammation-related proteins NF-κB,caspase-1,IL-1β,and IL-18 in lung tissue was significantly increased(P<0.05).Compared with the model group,SMXFZKF alleviated pathological changes such as airway inflammation in the lung tissues of PIC model rats,reduced serum levels of IL-1β,IL-6,TNF-α,and MDA,increased the contents of CAT and SOD,and down-regulated the expression of NF-κB,caspase-1,IL-1β,and IL-18 in lung tissues.Conclu-sion Xuanfei Zhike Formula may effectively alleviate coughing symptoms in PIC rats by down-regulating the expression of NF-κB,caspase-1,IL-1β,and IL-18,thereby inhibiting the secretion of inflammatory factors and alleviating pathological changes in lung tissues.
