为探究饲料中大豆卵磷脂对圆口铜鱼(Coreius guichenoti)肝脏营养代谢功能的调控机制,明确其在该鱼类饲料中的应用潜力,为圆口铜鱼规模化人工繁育提供理论支撑,本研究设置为期 8 周的养殖实验,采用大豆卵磷脂添加量为 0%(对照组)和 7%(处理组)的两种实验饲料饲喂圆口铜鱼后,比较两组实验鱼肝脏的脂质沉积状态差异;进一步通过肝脏组织转录组测序技术筛选两组间的差异表达基因,开展通路富集分析及脂代谢、糖代谢、氨基酸代谢相关关键基因的表达水平检测.结果显示:与对照组相比,7%大豆卵磷脂处理组显著改善了圆口铜鱼的肝脏脂质沉积状态.转录组测序分析显示,两组间的差异表达基因主要富集于脂肪酸β氧化、氨基酸代谢及糖异生等关键营养代谢通路.具体基因表达分析表明,处理组中脂代谢相关基因(adipor2、el)、糖代谢相关基因(g6pc1)及氨基酸代谢相关基因(spt、tod2)的表达水平均显著上调.本研究阐明了饲料中大豆卵磷脂调控圆口铜鱼肝脏营养代谢的潜在分子机制,证实了其改善肝脏脂质沉积、调控多代谢通路的作用效果,为大豆卵磷脂在圆口铜鱼饲料中的科学应用提供了直接理论依据.

This study systematically investigated the regulatory mechanism of dietary soy lecithin on liver nutri-tional metabolism of Coreius guichenoti.The effects of 0%(Ctrl)and 7%(PLs)soybean lecithin diet on liver me-tabolism of experimental fish were compared and analysed through an 8-week culture experiment.The results showed that compared with the control group,7%soybean lecithin diet(PLs)significantly improved the status of lipid deposition in the liver of Cupric fish.Transcriptome sequencing analysis of liver tissue revealed that dif-ferentially expressed genes were primarily focused in key metabolic pathways,including fatty acid β-oxidation,amino acid metabolism,and gluconeogenesis.Further analysis revealed that the expression levels of lipid metabo-lism-related genes(adipor2,el),glucose metabolism-related genes(g6pc1),and amino acid metabolism-related genes(spt,tod2)in the treatment group were significantly upregulated.Therefore,soy lecithin may play a role in the following ways:(1)activation of the fatty acid oxidation pathway to reduce lipid accumulation in the liver;(2)regulate the tryptophan metabolism pathway;(3)promote the process of glucose metabolism.This study provides a theoretical basis for the application of soybean lecithin in the feed of C.guichenoti,and has important practical significance for the large-scale artificial breeding of C.guichenoti.
