目的 探讨电针胃合募配穴是否通过调节磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3K)/蛋白激酶B(protein kinase B,AKT)/哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)信号通路改善抑郁模型小鼠的胃功能障碍.方法 选取30只雄性C57BL/6J小鼠,采用随机分组方式设立空白组、模型组和电针组,每组10只.通过慢性不可预知性温和应激构建抑郁症小鼠模型,电针组小鼠给予电针"足三里"和"中脘"穴,每日2次,持续7 d,空白组和模型组小鼠不进行任何干预.采用悬尾实验、强迫游泳实验、旷场实验、高架十字迷宫实验观察小鼠行为学变化,检测小鼠体质量、进食量、胃排空率、胃运动振幅,采用苏木精—伊红(hematoxylin-eo-sin,HE)染色法观察小鼠胃窦组织形态,采用TUNEL法检测小鼠胃窦组织平滑肌细胞凋亡率,采用Western blot法检测小鼠胃窦组织平滑肌中PI3K/AKT/mTOR通路蛋白表达水平.结果 电针干预能显著降低悬尾实验和强迫游泳实验中小鼠不动时间(P<0.05),增加旷场实验中小鼠进入中央区次数和运动总距离(P<0.05),以及高架十字迷宫中小鼠进入开放臂时间百分比和进入开放臂距离百分比(P<0.05),升高小鼠体质量、进食量、胃排空率和胃运动振幅(P<0.05),降低小鼠胃窦组织平滑肌细胞凋亡率(P<0.05),升高p-PI3K/PI3K、p-AKT/AKT、p-mTOR/mTOR比值(P<0.05).HE染色结果显示,电针组小鼠胃窦组织无明显病理变化.结论 电针可能通过激活PI3K/AKT/mTOR信号通路,抑制胃窦组织平滑肌细胞凋亡,改善抑郁症小鼠胃功能障碍.

Objective To investigate whether electroacupuncture at stomach He points and Front-Mu points improves gastric dysfunction in mice with depression by regulating the phosphoinositide-3 kinase(PI3K)/protein kinase-B(AKT)/mammalian target of rapamycin(mTOR)signaling pathway.Methods A total of 30 male C57BL/6 mice were randomly divided into blank group,model group,and electroacupuncture group,with 10 mice in each group,and chronic unpredictable mild stress(CUMS)was used to establish a mouse model of depression.The mice in the electroacupuncture group were given electroacupuncture at Zusanli and Zhongwan twice a day for 7 consecutive days,while those in the blank group and the model group were not given any intervention.The tail suspension test,the forced swimming test,the open field test,and the elevated plus maze test were used to observe behavioral changes;body weight,food intake,gastric emptying rate,and the amplitude of gastric motility were measured;HE staining was used to observe the histomorphology of gastric antrum,and TUNEL staining was used to observe the apoptosis of gastric antrum smooth muscle cells;Western blot was used to measure the expression levels of the proteins as-sociated with the PI3K/AKT/mTOR signaling pathway in gastric antrum smooth muscle.Results Electroacupuncture inter-vention significantly reduced immobility time in the tail suspension test and the forced swimming test(P<0.05)and increased the number of entries into the central zone and the total movement distance in the open field test(P<0.05).It also increased the percentage of open-arm dwell time(P<0.05)and the percentage of movement distance into the open arm(P<0.05),as well as body weight,food intake,gastric emptying rate,and the amplitude of gastric motility(P<0.05).In addition,it re-duced the apoptosis rate of gastric antrum smooth muscle cells(P<0.05)and increased the expression levels of the proteins as-sociated with the PI3K/AKT/mTOR signaling pathway(P<0.05).HE staining showed that there were no obvious pathologi-cal changes in the gastric antrum in the electroacupuncture group.Conclusion Electroacupuncture may improve gastric dys-function in mice with depression by activating the PI3K/AKT/mTOR signaling pathway and inhibiting the apoptosis of gastric antrum smooth muscle cells.
