外源赤霉素和低温对赤芍种子萌发和幼苗耐低温能力的影响OA
Effects of exogenous gibberellin and low temperature on the seed germination and the low temperature resistance of the seedlings of Paeonia lactiflora
为探究外源赤霉素(GA3)和低温对赤芍种子萌发和幼苗耐低温能力的影响,本试验以吉林(JL)、山东菏泽(SD)、河南洛阳(HN)3 个产地的赤芍为试验材料,测定常温、4℃低温处理下清水(CK)、外源GA3(300 mmol/L)浸种的赤芍种子萌发率、发芽时间,以及经常温、4℃低温处理以清水浸种并常温萌发的赤芍幼苗,在清水(CK)、100和300 mmol/L GA₃ 3种处理下的赤芍幼苗丙二醛(MDA)含量、过氧化氢酶(CAT)和过氧化物酶(POD)活性等.结果表明,与CK相比,300 mmol/L外源GA3能明显提高3个产地赤芍的生根率,缩短胚根和胚芽萌动的时间,并且提高发芽率和发芽势.300 mmol/L外源GA3能明显提升3个产地赤芍的CAT、POD活性,并且促进可溶性糖和可溶性蛋白积累;3个产地赤芍低温下CAT和POD活性均有所提升,可溶性糖和可溶性蛋白含量上升,以JL赤芍的抗氧化酶活性、可溶性糖和可溶性蛋白积累量最高;低温处理后3个产地赤芍的MDA含量均有所上升,以JL赤芍的MDA含量上升幅度最小.说明JL赤芍的耐低温能力较强,300 mmol/L外源GA3在一定程度上能提高赤芍的耐低温能力.
To investigate the effects of exogenous gibberellin(GA3)and low temperature on the seed germination and low temperature resistance of Paeonia lactiflora seeds,this experiment used P.lactiflora seeds from 3 locations-Jilin(JL),Shandong(SD),and Henan(HN)as the experimental materials.The germination rate and germination time of the P.lactiflora seeds after soaking in water(CK)and with exogenous GA3(300 mmol/L)at room temperature and 4℃low temperature were determined.As well as the P.lactiflora seedlings that were frequently treated at room temperature and 4℃low temperature,soaked in water for seed germination and then germinated at room temperature.The contents of malondialdehyde(MDA),the activities of catalase(CAT)and peroxidase(POD)in the young rhubarb seedlings were measured under 3 treatments.water(CK),100 and 300 mmol/L GA₃.The results showed that,compared with CK,300 mmol/L exogenous GA3 could significantly increase the rooting rate of 3 production areas of P.lactiflora,shorten the germination time of embryonic roots and embryos,and also enhance the germination rate and germination potential.300 mmol/L exogenous GA3 could significantly enhance the CAT and POD activities of 3 production areas of P.lactiflora,and promote the accumulation of soluble sugars and soluble proteins;the CAT and POD activities of 3 production areas of P.lactiflora under low temperature all increased,as well as the contents of soluble sugars and soluble proteins.Among them,the antioxidant enzyme activity,soluble sugar and soluble protein accumulation of JL P.lactiflora were the highest;after low-temperature treatment,the MDA content of P.lactiflora from the 3 production areas all increased,and the MDA content of JL P.lactiflora showed the smallest increase.This indicated that JL P.lactiflora had a stronger ability to withstand low temperature,and 300 mmol/L exogenous GA3 could to some extent enhance the cold resistance of P.lactiflora.
