海洋产尿酸氧化酶菌株的发酵优化及发酵放大OA
Fermentation optimization and fermentation scale-up of marine Urate oxidase-producing bacterial strains
为提高产尿酸氧化酶菌株迈索尔谷氨酸杆菌(Arthrobacter mysorens)Dw-17a的产酶能力,本试验采用响应面法对Dw-17a发酵产酶条件进行优化,并进行5L规模的发酵放大试验.结果显示:最适发酵条件为温度15.6℃、pH 8.1、装液量 95 mL/500 mL、转速 160 r/min、接种量 3%.在此条件下平均酶活可达到 58.24 U/mL,比优化前提高了 23.3%.在此基础上进行 5L发酵罐放大试验,结果表明 5L规模下的最适发酵条件为葡萄糖 2%、酵母浸粉 1.5%、转速 150 r/min、装液量 40%、接种量 3%.在此条件下平均酶活可达到 42.11 U/mL.
In order to improve the enzyme production capacity of Urate oxidase-producing strain Arthrobacter mysorens Dw-17a.Response surface methodology was used to optimize the enzyme-producing conditions for the fermentation of Dw-17a and fermentation scale-up experiments were carried out at a scale of 5 L.The results showed that the optimum fermentation conditions were temperature 15.6℃,pH 8.1,liquid filling amount 95 mL/500 mL,rotation speed 160 r/min and inoculum volume 3%.The average enzyme activity could reach 58.24 U/mL under this condition,which was 23.3%higher than that before optimization.On the basis of this experiment,a 5 L fermenter was scaled up,and the results showed that the optimal fermentation conditions for the 5 L scale were glucose 2%,yeast extract powder 1.5%,rotation speed 150 r/min,liquid filling amount 40%,and inoculum volume 3%,and the average enzyme activity could reach 42.11 U/mL under these conditions.
