羊驼源枯草芽孢杆菌分离培养与鉴定OA
Isolation,cultivation,and identification of Bacillus subtilis originating from alpaca
为得到稳定性高、抗逆性强、高蛋白酶活性、高淀粉酶活性、低耐药性的枯草芽孢杆菌,本试验从健康羊驼的新鲜粪便中分离、纯化得到 3 株枯草芽孢杆菌菌株,并进行形态学鉴定、16S rRNA测序、蛋白酶试验、淀粉酶试验、耐受性试验及药敏试验,以确定菌株种类及功能.结果表明:筛选得到的3株菌革兰氏染色为紫色杆状,经16S rRNA测序和序列比对确定为枯草芽孢杆菌,编号分别为 21K、25K、29K.其中 29K整体耐受特性最好,在 3 株菌株中呈现高蛋白酶活性、高淀粉酶活性和低耐药性.综上所述,29K作为枯草芽孢杆菌益生菌制剂潜力较高.
In order to obtain Bacillus subtilis strains with high stability,strong stress resistance,high protease activity,high amylase activity,and low drug resistance,three strains of Bacillus subtilis were isolated and purified from the fresh feces of healthy alpacas in this study.Subsequently,morphological identification,16S rRNA sequencing,protease tests,amylase tests,tolerance tests,and drug sensitivity tests were conducted to determine the species and functions of the strains.The results showed that the three selected strains were Gram-positive purple rods and were identified as Bacillus subtilis through 16S rRNA sequencing and sequence alignment,with the strain numbers 21K,25K,and 29K,respectively.Further experiments indicated that 29K exhibited the best overall tolerance characteristics among the three strains,showing high protease activity,high amylase activity,and low drug resistance.In summary,29K had high potential as a probiotic preparation of Bacillus subtilis.
