枯草芽孢杆菌DF-54的产酶优化及其对烤制晒烟的提质效果研究OA
Study on the optimization of enzyme production by Bacillus subtilis DF-54 and its effect on the quality improvement of baked sun-cured tobacco
为降低烤制晒烟的淀粉含量,增强新型原料工业可塑性与安全性.本研究利用响应面法和正交试验,对一株改善烤制晒烟品质的枯草芽孢杆菌DF-54进行产酶培养基和产酶条件优化,对优化前后菌株所产粗酶液以及无菌水发酵烟叶的常规化学成分和挥发性物质进行检测分析.结果表明,菌株DF-54的最佳产酶培养基配方为:麦麸3.50 g/L,豆粕20.41 g/L,KCl 4.64 g/L;最佳产酶条件为:接种量2%,转速200 rpm,时间36 h.与基础培养基和产酶条件相比,淀粉酶酶活力提升率为103.9%.烟叶经发酵后,总糖含量增加16.45%,还原糖含量增加20.76%,淀粉降解率为17.21%,巨豆三烯酮、香叶基丙酮、β-紫罗兰酮和茄尼酮等中性致香成分含量显著增加;香叶基丙酮和脱氢二氢紫罗兰酮等物质与淀粉酶、果胶酶、木聚糖酶和纤维素酶呈显著正相关,与中性、碱性蛋白酶呈显著负相关.本研究结果为烟草源微生物提高烟叶品质的工业化应用提供理论基础.
To reduce the starch content in flue-cured sun-dried tobacco and enhance the indus-trial processability and safety of novel tobacco materials,this study employed response sur-face methodology and orthogonal experimental design to optimize the enzyme-producing cul-ture medium and fermentation conditions for Bacillus subtilis DF-54,a strain that improves the quality of flue-cured sun-dried tobacco.Comparative analyses were conducted on the crude enzyme solutions produced by the strain under both optimized and non-optimized condi-tions,as well as on the routine chemical components and volatile compounds of semi-finished tobacco products fermented with these enzyme solutions versus sterile water.The results demonstrated that the optimal enzyme-producing medium composition for strain DF-54 was:wheat bran 3.50 g/L,soybean meal 20.41 g/L,KCl 4.64 g/L.The optimal fermentation conditions were:inoculum size 2%,agitation speed 200 rpm,and fermentation time 36 h.Compared to the basal medium and initial fermentation conditions,amylase activity increased by 103.9%.After fermentation,tobacco leaves exhibited a 16.45%increase in total sugar content,a 20.76%increase in reducing sugar content,and a starch degradation rate of 17.21%.The contents of key neutral aroma components,including megastigmatrienone,geranylacetone,β-ionone,and solanesone,were significantly elevated.Geranylacetone and dehydrodihydro-β-ionone showed significant positive correlations with amylase,pectinase,xylanase,and cellulase activities,while exhibiting significant negative correlations with neu-tral and alkaline protease activities.These findings provide a theoretical foundation for the industrial application of tobacco-derived microorganisms in enhancing tobacco leaf quality.
