结直肠癌组织中YTHDF1 mRNA和MGP mRNA表达与免疫相关基因的相关性及预后价值分析OA
Correlation between YTHDF1 mRNA,MGP mRNA Expression and Immune Related Genes in Colorectal Cancer and Their Prognostic Value
目的 研究结直肠癌(CRC)组织中YTH结构域N6-甲基腺嘌呤RNA结合蛋白1(YTHDF1)mRNA、基质GLA蛋白(MGP)mRNA表达与免疫相关基因表达的相关性及预后价值分析.方法 选取2019年2月~2021年2月北京中医医院内蒙古医院诊治的98例CRC患者,采用实时荧光定量PCR和免疫组化检测组织中YTHDF1、MGP的mRNA和蛋白表达.Pearson相关分析YTHDF1 mRNA、MGP mRNA与程序性死亡因子1(PD-1)mRNA、程序性死亡因子1配体(PD-L1)mRNA及白细胞介素6(IL-6)mRNA的相关性.Kaplan-Meier曲线及COX回归分析YTHDF1 mRNA、MGP mRNA对CRC预后的影响.结果 CRC癌组织中YTHDF1蛋白(91.84%vs 7.14%),MGP蛋白(89.80%vs 11.22%)阳性率高于癌旁组织,差异具有统计学意义(χ2=140.606、121.013,均P<0.05).相比于癌旁组织,CRC癌组织中YTHDF1 mRNA(3.11±0.41 vs 1.03±0.22)、MGP mRNA(2.89±0.38 vs 0.76±0.20)、PD-1 mRNA、PD-L1 mRNA及IL-6 mRNA表达量升高(t=26.277~49.103,均P<0.05).CRC癌组织中YTHDF1 mRNA、MGP mRNA与免疫相关基因PD-1 mRNA、PD-L1 mRNA、IL-6 mRNA表达呈正相关(r=0.667~0.775,均P<0.05).TNM分期Ⅲ期、淋巴结转移患者的CRC组织中YTHDF1 mRNA、MGP mRNA表达高于TNM分期Ⅰ~Ⅱ期及无淋巴结转移患者,差异具有统计学意义(t=18.825~30.469,均P<0.05).YTHDF1 mRNA高表达组和低表达组三年生存率分别为45.83%(22/48)、72.00%(36/50),差异具有统计学意义(Log-rank χ2=8.946,P=0.003).MGP mRNA高表达组和低表达组三年生存率分别为42.55%(20/47)、74.51%(38/51),差异具有统计学意义(Log-rank χ2=12.500,P<0.001).多因素COX回归分析,YTHDF1 mRNA(OR=1.384,95%CI=1.088~1.761)、MGP mRNA(OR=1.324,95%CI=1.080~1.624)、TNM分期Ⅲ期和淋巴结转移是影响CRC预后的危险因素.结论 CRC组织中YTHDF1、MGP的mRNA和蛋白表达升高,与免疫相关基因及不良临床病理特征有关,是评估CRC预后的肿瘤标志物.
Objective To study the expression of YTH N6-methyladenosine RNA binding protein 1(YTHDF1)mRNA and Matrix Gla protein(MGP)mRNA in colorectal cancer(CRC)tissues and explore their correlation with immune-related genes and prog-nostic value.Methods A total of 98 patients with CRC diagnosed and treated in Beijing Traditional Chinese Medicine Hospital Inner Mongolia Hospital from February 2019 to February 2021 were selected.Real-time fluorescence quantitative PCR and im-munohistochemistry were used to detect the expression of YTHDF1 and MGP mRNA and protein in tissues.The correlation be-tween YTHDF1 mRNA,MGP mRNA and Programmed death factor 1(PD-1)mRNA,Programmed death factor 1 ligand(PD-L1)mRNA,and interleukin-6(IL-6)mRNA were analyzed by pearson correlation analysis.Kaplan-Meier curve and COX regression were used to analyze the effect of YTHDF1 mRNA and MGP mRNA on the prognosis of CRC.Results The positive rates of YT-HDF1 protein(91.84%vs 7.14%)and MGP protein(89.80%vs 11.22%)in CRC cancer tissues were significantly higher than those in adjacent tissues,the differencse were statistically significant(χ2=140.606,121.013,all P<0.05).Compared with adjacent tissues,the expression of YTHDF1 mRNA(3.11±0.41 vs 1.03±0.22),MGP mRNA(2.89±0.38 vs 0.76±0.20),PD-1 mRNA,PD-L1 mRNA and IL-6 mRNA in CRC cancer tissues were significantly higher(t=26.277~49.103,all P<0.05).There was a posi-tive correlation between YTHDF1 mRNA,MGP mRNA and immune-related genes PD-1 mRNA,PD-L1 mRNA and IL-6 mRNA expression in CRC cancer tissues(r=0.667~0.775,all P<0.05).The expression of YTHDF1 mRNA and MGP mRNA in CRC tis-sues of patients with TNM stage III and lymph node metastasis was significantly higher than that of patients with TNM stage I~II and no lymph node metastasis,the differencse were statistically significant(t=18.825~30.469,all P<0.05).The 3-year survival rate of YTHDF1 mRNA high expression group and low expression group was 45.83%(22/48),72.00%(36/50),and the difference was statistically significant(Log-rank χ2=8.946,P=0.003).The 3-year survival rate of MGP mRNA high expression group and low expression group was 42.55%(20/47),74.51%(38/51),and the difference was statistically significant(Log-rank χ2=12.500,P<0.001).Multivariate COX regression anslysis,YTHDF1 mRNA(OR=1.384,95%CI=1.088~1.761),MGP mRNA(OR=1.324,95%CI=1.080~1.624),TNM stage III and lymph node metastasis were risk factors affecting the prognosis of CRC.Conclusions The elevated mRNA and protein expression of YTHDF1 and MGP in CRC are associated with immune related genes and adverse clinical pathological features.Both of them are tumor markers for evaluating CRC prognosis.
