2022年2月,山东省某规模化白羽肉鸡场鸡群发生以严重肾脏损伤为特征的传染病,发病原因不明.为明确其病因,本试验采集病料,经RT-PCR检测、病毒分离和S1基因测序分析,确认分离到一株致肾病变型鸡传染性支气管炎病毒(IBV)毒株,并将其命名为RP-NR22,S1基因的遗传进化分析表明,该毒株属于GI-19谱系.为深入了解该毒株的致病特征,将该毒株接种SPF鸡进行动物回归试验,致病性结果证实,RP-NR22可导致SPF鸡100%(10/10)产生肾脏病变,并有40%(4/10)的致死率.值得注意的是,目前广泛使用的商品化IBV ELISA试剂盒无法检测到感染鸡体内抗体的产生.本研究通过分离到一株具有较强致病性的肾型IBV,为进一步研究肾型IBV的致病机理及科学防控提供了基础素材和数据.

During February 2022,a contagious illness leading to significant kidney impairment was identified in a flock of white-feathered broiler chickens at a large farm in Shandong Province,China.However,the specific etiological factors remain unclear.In order to ascertain the causative agent of the disease,samples were gathered for RT-PCR testing,virus isolation,and S1 gene sequencing analysis.A nephropathogenic strain of avian infectious bronchitis virus(IBV)was isolated and designated as RP-NR22.Through analysis of genetic evolution centered on the S1 gene,it was determined that this strain is part of the GI-19 lineage and showcases mutations at critical glycosylation sites.In order to explore the pathogenic features of RP-NR22,the virus was administered to SPF chickens to evaluate its pathogenicity.Animal experiments have validated that RP-NR22 induced kidney lesions in all SPF chickens(10/10)and resulted in a mortality rate of 40%(4/10).The commercially available IBV ELISA kits commonly used at present have shown an inability to identify antibody responses in chickens infected with the virus.This report marks the initial identification of a nephropathogenic IBV strain isolated from both domestic and international sources.The findings of this study offer foundational resources and information for advancing research on the pathogenic pathways of nephropathogenic IBV and the development of effective preventive and control measures.
