目的 该研究从"肺与大肠相表里"出发结合现代医学中"肠-肺轴"理论,探讨桑皮止咳方对呼吸道合胞病毒(RSV)感染导致的咳嗽小鼠模型的治疗作用,同时分析其对肠道微生物及短链脂肪酸(SCFAs)水平的调控作用.方法 该研究选取60只雌性BALB/c小鼠,实验组包括:空白对照组、RSV感染模型组、西药治疗组,以及桑皮止咳方进行低、中、高剂量治疗组,各组包含10只小鼠.除空白对照组外,其他组小鼠均通过RSV滴鼻以建立咳嗽模型,并通过辣椒素雾化进一步诱导咳嗽反应.记录小鼠一般状态,并利用HE染色对其肺肠组织的形态学改变进行评估.此外,为深入分析肠道菌群变化,收集盲肠的粪便样本,通过选择培养基定量分析其中的肠球菌、乳酸杆菌及双歧杆菌的数量,并运用气相色谱法测定粪便中常见SCFAs的含量.结果 与空白组相比较,模型组小鼠出现明显呼吸道疾病症状以及肺、肠组织的病理变化,肠道菌群水平显著下降(P<0.01,P<0.05),相应代谢产物SCFAs的水平显著下降(P<0.01,P<0.05);与模型组相比较,治疗组的肺肠组织形态学改变整体好转,相应一般状态也较模型组有明显改善,并且在应用桑皮止咳方后肠道菌群以及代谢产物SCFAs均一定程度上趋近于正常组.结论 桑皮止咳方不仅能减轻小鼠咳嗽等呼吸道症状,还可以调控模型小鼠的肠道菌群丰度,使其向正常水平恢复.该方通过肺肠同治法治疗感染后咳嗽,该研究为桑皮止咳方在"肺肠同治"儿童RSV感染后咳嗽治疗中的应用提供了科学依据.

Objective In this study,we investigated the therapeutic effect of Sangpi Zhike Decoction(桑皮止咳方)on a mouse model of cough caused by respiratory syncytial virus(RSV)infection,and analyzed its regulatory effect on intestinal microbes and short-chain fatty acids(SCFAs)levels based on the"lung and large intestine phases"and the theory of"gut-lung axis"in modern medicine.Methods In this study,60 female BALB/c mice were selected,and the experimental groups included blank control group,RSV infection model group,western medicine treatment group,and Sangpi Zhike Decoction low,medium and high dose treatment groups,each group included 10 mice.Except for the blank control group,the mice in the other groups were treated with RSV nasal drops to establish a cough model,and the cough response was further induced by capsaicin atomization.The general state of mice was recorded,and the morphological changes of lung and intestinal tissues were evaluated by HE staining.In addition,in order to deeply analyze the changes of intestinal microbiota,fecal samples were collected from the cecum,and the number of enterococci,lactobacilli and bifidobacteria in the group was quantitatively analyzed by selecting the medium,and the content of common SCFAs in the feces was determined by gas chromatography.Results Compared with the blank group,the mice in the model group showed obvious symptoms of respiratory tract diseases and pathological changes in lung and intestinal tissues.The level of intestinal flora decreased significantly(P<0.01,P<0.05)and the level of corresponding metabolites SCFAs decreased significantly(P<0.01,P<0.05).Conclusion Sangpi Zhike Decoction can not only alleviate respiratory symptoms such as cough in mice,but also regulate the gut microbiota abundance in model mice,restoring it to normal levels.This decoction treats post-infection cough through lung-intestine coordinated therapy.This study provides a scientific basis for the application of Sangpi Zhike Decoction in treating post-RSV infection cough in children under the"lung-intestine co-treatment"approach.
