水晶兰苷对APP-N2a细胞神经保护作用及机制研究OA
Neuroprotective Effect and Mechanism of Monotropein on APP-N2a Cells
目的 探讨水晶兰苷(Mon)对携带瑞典突变型人淀粉样前体蛋白(APPswe)基因腺病毒感染的小鼠神经母细胞瘤细胞(N2a)的神经保护作用及作用机制.方法 利用携带APPswe基因的腺病毒感染N2a细胞建立AD细胞模型;利用CCK-8法筛选Mon及核因子-E2相关因子2(Nrf2)激动剂(Nrf2-Ac)的最佳给药浓度;利用流式细胞技术法检测Mon对APP-N2a细胞凋亡的影响;利用ELISA法检测Mon对APP-N2a细胞相关炎症因子表达的影响;利用免疫细胞化学法检测Mon对APP-N2a细胞Nrf2、血红素加氧酶1(HO-1)蛋白表达的影响.结果 20μM Mon和1000 nM Nrf 2-Ac能显著提高APP-N2a细胞存活率;Mon及Nrf 2-Ac治疗后能显著降低APP-N2a细胞凋亡率,降低白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子α(TNF-α)等炎症因子表达,并有效提高APP-N2a细胞Nrf2、HO-1蛋白表达.结论 Mon对APP-N2a细胞具有神经保护作用,其机制与激活Nrf2/HO-1信号通路有关.
Objective To investigate the neuroprotective and mechanism of monotropein(Mon)on the N2a cell infected by adenoviral carrying the APPswe gene.Methods The CCK-8 assay was used to detect the optimal concentration by Mon and Nrf2 activator(Nrf2-Ac)on APP-N2a cells,flow cytometry was used to detect the effect of Mon and Nrf2-Ac on apoptosis of APP-N2a cells,ELISA assay was used to detect the effect of Mon and Nrf2-Ac on inflam-cytokines of APP-N2a cells and immunohistochemistry assay was used to detect the effect of Mon and Nrf2-Ac on Nrf2,HO-1 protein expression of APP-N2a cells.Results The results showed that 20 μM Mon and 1000 nM Nrf2-Ac could increase the survival rate and reduce apoptosis of the APP-N2a cells.The levels of IL-6,IL-1β,TNF-α were decreased while the level of Nrf2,HO-1 protein expression was increased.Conclusion Mon has the most significant neuroprotective in the APP-N2a cells by activating the Nrf2/HO-1 signaling pathway.
