Recent Advances in Research on Histidine Triad Superfamily Proteins

组氨酸三联体(histidine triad,HIT)蛋白广泛存在于不同的生物体中,是一类核苷酸水解酶/转移酶超家族,其特征性催化基序为HxHxH,主要包括HINT、FHIT、GALT三大分支.组氨酸三联体超家族蛋白在DNA修复、信号转导及疾病进程中扮演关键角色,其异常表达或突变也与多种疾病的发生有关,如癌症、神经退行性疾病等.因此,对组氨酸三联体超家族蛋白的结构特点及催化机制进行系统性的梳理比较,可以为理解该家族蛋白在不同生理过程中的作用机制提供帮助,也可以为基于该家族蛋白的小分子药物设计提供参考,并最终服务于癌症等疾病的早期诊断和治疗.

The histidine triad(HIT)proteins are widely distributed across various organisms and constitute a superfamily of nucleotide hydrolases/transferases,characterized by the catalytic motif HxHxH.These proteins can be classified into three major branches:HINT,FHIT,and GALT.HIT superfamily proteins play crucial roles in DNA repair,signal transduction,and disease progression,and their aberrant expression or mutation is also associated with various diseases,such as cancer and neurodegenerative diseases.Therefore,a systematic comparison and analysis of the structural features and catalytic mechanisms of the HIT superfamily proteins is essential for under-standing their functional roles in different physiological processes.This knowledge may provide valu-able insights for the design of small-molecule drugs targeting these proteins and ultimately contribute to the early diagnosis and treatment of diseases such as cancer.
