Effects of Different Needle Retention Times of Electroacupuncture on GABAB Receptor Expression in the Prefrontal Cortex of Rats with Traumatic Brain Injury-Induced Coma

目的:观察电针不同留针时间对脑外伤昏迷大鼠前额叶皮质γ-氨基丁酸B(GABAB)受体表达的影响,并探讨电针促醒的潜在神经作用机制.方法:24只清洁级大鼠依据随机数字表随机分为空白组、模型组、电针组A及电针组B,每组各6只.其中空白组不作处理,其余各组大鼠采用自由落体撞击法建立脑外伤昏迷模型大鼠.造模成功后,电针组A和电针组B针刺百会、水沟穴后分别给予电针刺激20 min和30 min,模型组仅针刺,空白组大鼠不作任何处理,1次/d,连续7 d.观察大鼠行为学变化情况,取前额叶皮质组织采用Western blot检测前额叶皮质GABAB、水通道蛋白-4(AQP-4)及丙二醛(MDA)蛋白的表达情况.结果:与空白组比较,模型组前额叶皮质GABAB受体、AQP-4和MDA蛋白表达量极显著上升(P<0.001).与模型组比较,电针组A的AQP-4和MDA蛋白表达量显著下降(P<0.01);电针组B的AQP-4和MDA蛋白表达量极显著下降(P<0.001);电针组A和电针组B的GABAB受体蛋白表达量均显著下降(P<0.01).与电针组A比较,电针组B的GABAB受体、AQP-4和MDA蛋白表达有下降趋势,但差异无统计学意义.结论:GABAB受体是电针促醒作用的重要神经递质,电针促醒的分子机制可能与下调GABAB受体的表达有关,随着留针时间增加,电针对下调GABAB受体的表达未产生明显变化.

Objective:To observe the effects of different needle retention times of electroacupuncture on the expression of gamma-aminobutyric acid B(GABAB)receptors in the prefrontal cortex of rats with traumatic brain injury-induced coma,and to explore the potential neural mechanism of electroacupuncture in promoting arousal.Methods:Twenty-four clean-grade rats were randomly divided into a blank group,a model group,electroacupuncture group A,and electroacupuncture group B according to a random number table,with 6 rats in each group.The blank group received no treatment.Rats in the other groups were used to establish a traumatic brain injury-induced coma model using the free-fall impact method.After successful modeling,electroacupuncture group A and electroacupuncture group B received electroacupuncture stimulation at Baihui(GV20)and Shuigou(GV26)acupoints for 20 minutes and 30 minutes respectively,following needle insertion.The model group only received needle insertion,and the blank group received no treatment.The intervention was administered once daily for 7 consecutive days.Behavioral changes in the rats were observed.Prefrontal cortex tissue was harvested,and Western blot was used to detect the protein expression of GABAB,aquaporin-4(AQP-4),and malondialdehyde(MDA)in the prefrontal cortex.Results:Compared with the blank group,the protein expression levels of GABAB receptors,AQP-4,and MDA in the prefrontal cortex of the model group were significantly increased(P<0.001).Compared with the model group,the protein expression levels of AQP-4 and MDA in electroacupuncture group A were significantly decreased(P<0.01);the protein expression levels of AQP-4 and MDA in electroacupuncture group B were extremely significantly decreased(P<0.001);the protein expression levels of GABAB receptors in both electroacupuncture group A and electroacupuncture group B were significantly decreased(P<0.01).Compared with electroacupuncture group A,electroacupuncture group B showed a decreasing trend in the protein expression of GABAB receptors,AQP-4,and MDA,but the difference was not statistically significant.Conclusion:The GABAB receptor is an important neurotransmitter involved in the arousal-promoting effect of electroacupuncture.The molecular mechanism of electroacupuncture promoting arousal may be related to the down-regulation of GABAB receptor expression.No additional effect on the down-regulation of GABAB receptor expression was observed with prolonged electroacupuncture needle retention time.
