生、制木香制备香连散对腹泻小鼠水通道蛋白和Na+-K+-ATP酶的影响OA
Effects of Xianglian Powder Prepared from Raw and Processed Aucklandiae Radix on Aquaporins and Na+-K+-ATPase in Diarrhea Mice
目的:比较生、制木香制备的香连散抗腹泻的药效学差异及作用机制.方法:选取SPF级雄性昆明小鼠56只,随机分为空白组、模型组、生高组(0.771 4 g/mL)、生低组(0.385 7 g/mL)、制高组(0.771 4 g/mL)、制低组(0.385 7 g/mL)、阳性组(盐酸洛哌丁胺2 mg/kg).除空白组外,其余各组小鼠连续灌胃番泻叶(0.1 mL/10 g)7 d,造模成功后,再连续4 d给予相应药物治疗,同时在给药结束0.5 h后用番泻叶巩固腹泻模型.造模后置于代谢笼内,记录总便数、稀便数及稀便等级并计算稀便率、单鼠平均稀便级、腹泻率及腹泻指数.禁食不禁水12 h,于第11天给药结束后取血,采用酶联免疫吸附法(ELISA)检测血清中Na+-K+-ATP酶活性、水通道蛋白AQP3、AQP4表达水平.结果:制高组小鼠平均腹泻指数(0.29±0.3)、平均腹泻率47.8%,与模型组相比显著降低(P<0.05);制高组Na+-K+-ATP酶活性上升(17.60±2.50),与模型组相比显著升高(P<0.05);制高组小鼠血清中水通道蛋白AQP3的表达水平上调(156.45±6.39),与模型组相比显著升高(P<0.01),水通道蛋白AQP4的表达水平上调(715.24±73.44),与模型组相比显著升高(P<0.001);与制高组相比,第3天生低组腹泻指数有显著性差异(P<0.05)、生低组血清中Na+-K+-ATP酶活性均呈现显著性降低(P<0.05)、生高组AQP4的表达水平上调显著(P<0.05).结论:高剂量的制香连散组止泻作用优于生香连散组,并可能通过提高Na+-K+-ATP酶活性水平,上调水通道蛋白AQP3、AQP4的表达水平而达到止泻作用.
Objective:To compare the pharmacodynamic differences and mechanisms of action of Xianglian Powder prepared from raw and processed Aucklandiae Radix against diarrhea.Methods:Fifty-six SPF-grade male Kunming mice were randomly divided into a blank group,a model group,a raw-high-dose group(0.771 4 g/mL),a raw-low-dose group(0.385 7 g/mL),a processed-high-dose group(0.771 4 g/mL),a processed-low-dose group(0.385 7 g/mL),and a positive group(loperamide hydrochloride 2 mg/kg).Except for the blank group,mice in the other groups were administered senna leaf decoction(0.1 mL/10 g)by gavage for 7 consecutive days to establish the model.After successful modeling,corresponding drug treatments were administered for 4 consecutive days.Simultaneously,0.5 hours after the last administration,the diarrhea model was reinforced using senna leaf.After modeling,mice were placed in metabolic cages to record the total number of feces,the number of loose stools,and the loose stool grade,and to calculate the loose stool rate,average loose stool grade per mouse,diarrhea rate,and diarrhea index.After fasting for 12 hours with free access to water,blood was collected on the 11th day after the final administration.Serum Na+-K+-ATPase activity and the expression levels of aquaporins AQP3 and AQP4 were detected by enzyme-linked immunosorbent assay(ELISA).Results:The average diarrhea index(0.29±0.3)and average diarrhea rate(47.8%)in the processed-high-dose group were significantly lower compared to the model group(P<0.05).Na+-K+-ATPase activity increased in the processed-high-dose group(17.60±2.50),which was significantly higher than those in the model group(P<0.05).The expression level of aquaporin AQP3 in the serum of mice in the processed-high-dose group was up-regulated(156.45±6.39),significantly higher than in the model group(P<0.01).The expression level of aquaporin AQP4 was up-regulated(715.24±73.44),significantly higher than in the model group(P<0.001).Compared with the processed-high-dose group,the diarrhea index on the third day showed a significant difference in the raw-product groups(P<0.05);serum Na+-K+-ATPase activity in the raw-low-dose group showed a significant decrease(P<0.05);and the expression level of AQP4 in the raw-high-dose group was significantly up-regulated(P<0.05).Conclusion:The antidiarrheal effects of the high-dose processed Xianglian Powder groups are superior to that of the raw Xianglian Powder groups.Its mechanism may involve increasing Na+-K+-ATPase activity levels and up-regulating the expression levels of aquaporins AQP3 and AQP4 to achieve the antidiarrheal effect.
