毛相思子黄酮类生物合成关键酶基因的筛选及鉴定OA北大核心
Screening and Identification of Key Enzyme Genes for Flavonoid Biosynthesis in Abrus mollis
目的:筛选和鉴定毛相思子黄酮类成分合成途径中的关键酶基因,为充分利用毛相思子药用资源提供必要的理论依据.方法:通过代谢组学分析和鉴定毛相思子叶和茎中黄酮类的差异成分,利用转录组测序和生物信息学数据库进行通路富集分析,筛选出与黄酮生物合成相关的关键基因.结果:通过代谢组学分析发现,毛相思子的叶和茎中共检测出196 个含量有差异的黄酮类成分,茎有47 种成分比叶的含量高,叶有149 种茎的含量高,毛蕊异黄酮、木犀草素、芒柄花素等活性成分在茎和叶中含量差异明显.转录组分析表明,不同部位比较组中PAL、4CL、C4H、HID等 13 种合成酶共同调控 32 个黄酮类成分.经过qRT-PCR结果显示,随机选取的 8 个参与黄酮合成的酶基因在毛相思子叶和茎中表达趋势与转录组测序结果一致.结论:毛相思子茎、叶部位中毛蕊异黄酮等黄酮类成分含量具有明显差异,其合成途径中关键酶基因的差异表达对毛相思子中黄酮类成分具有显著的调控作用,该研究结果对于日后毛相思子不同部位的合理利用具有重要的指导意义.
Objective:To screen and identify the key enzyme genes in the flavonoid component synthesis pathway of Abrus mollis,so as to provide a necessary theoretical basis for the full utilization of the medicinal resources of Abrus mollis.Methods:Metabolomics was used to analyze and identify the differential components of flavonoids in the leaves and stems of Abrus mollis.Pathway enrichment a-nalysis was conducted using transcriptome sequencing and bioinformatics databases to screen out the key genes related to flavonoid bio-synthesis.Results:Through metabolomics analysis,a total of 196 flavonoids with different contents were obtained in the leaves and stems of Abrus mollis.Among them,the concent of 47 compounds in the stems were higher than those in the leaves,and 149 compounds con-tents in the leaves were higher than those in the stems.The contents of active components such as calycosin,luteolin,formononetin in the stems and leaves were significantly different.Transcriptome analysis indicated that in the comparison groups of different parts,13 syn-thetic enzymes such as PAL,4CL,C4H,HID jointly regulated 32 flavonoid components.The results of qRT-PCR showed that the expres-sion trends of the 8 randomly selected enzyme genes involved in flavonoid synthesis in the leaves and stems of Abrus mollis were consist-ent with the results of transcriptome sequencing.Conclusion:There are significant differences in the contents of flavonoid components such as calycosin in the stems and leaves of Abrus mollis.The differential expression of key enzyme genes in their synthetic pathways has a significant regulatory effect on the flavonoid components in Abrus mollis.The research results have important guiding significance for the rational utilization of different parts of Abrus mollis in the future.
