特异性PCR方法鉴定大黄䗪虫丸中的水蛭成分OA北大核心
Authentication of Hirudo in Dahuang Zhechong Pills by Specific PCR Method
目的:建立特异性PCR方法,解决中成药大黄䗪虫丸中水蛭成分物种真实性的鉴定问题.方法:根据宽体金线蛭(WP)、菲牛蛭(PM)、日本医蛭(HN)、光润金线蛭(WL)的线粒体全基因序列差异设计引物;采用十六烷基三甲基溴化铵(CTAB)、酚仿抽提法提取与纯化总DNA,以此为模板建立聚合酶链反应(PCR)并进行了反应条件的筛选和优化,同时考察该方法的特异性和灵敏度,并对市售大黄䗪虫丸中的水蛭成分进行了实测.结果:设计所得 4 对特异性引物PWP、PHN、PPM及PWL,分别在 63、154、155、197 bp处扩增出清晰的目标条带,且未观察到有非特异性条带的干扰;4 对特异性引物的检测限均为 1.0 ng/μL;可行性验证结果表明,10 批市售大黄䗪虫丸中有 3批仅检出宽体金线蛭DNA,被鉴定为合格产品;其余 7 批则未检测出目标条带.结论:该研究所建立的检测方法具有良好的特异性和灵敏度,可用于鉴定经典名方大黄䗪虫丸中应含的水蛭成分,为含水蛭成方制剂的市场监管和基原追溯等工作提供了新的检测手段.
Objective:To establish a specific PCR method for the identification of species authenticity of Hirudo in Dahuang Zhe-chong Pills,a Chinese patent medicine.Methods:Primers were designed based on the differences in the complete mitochondrial gene se-quences of Whitmania pigra(WP),Poecilobdella manillensis(PM),Hirudo nipponica(HN)and Whitmania laevis(WL).Total DNA was extracted and purified by cetyltrimethylammonium bromide(CTAB)and phenol mimesis.Based on this template,polymerase chain reaction(PCR)was established and the reaction conditions were screened and optimized.The specificity and sensitivity of the method were investigated,and the Hirudo components in commercially available Dahuang Zhechong Pills were measured.Results:The four pairs of specific primers PWP,PHN,PPM and PWL obtained from the design amplified clear target bands at 63,154,155 and 197 bp,re-spectively,and no interference from non-specific bands was observed.The detection limits of the four pairs of specific primers were all 1.0 ng/μL.Feasibility verification results showed that three out of ten commercially available batches of Dahuang Zhechong Pills were identified as qualified products with only DNA of Whitmania pigra.The target bands were not detected in the remaining 7 batches.Con-clusion:The method established in this study has good specificity and sensitivity,which can be used to identify Hirudo in the classic prescription Dahuang Zhechong Pills,providing a new detection method for the market regulation and origin tracing of Hirudo-containing formulations.
