目的:对铁皮石斛种苗工厂化生产工艺进行优化,以降低种苗生产成本,提高移栽成活率.方法:将铁皮石斛种子液体悬浮培养 30 d,待种子变为原球茎后吸取50 粒,可视化点播到不同配方培养基,培养120 d后以试管苗生长势为评判标准,筛选最佳培养基;生根阶段用椰糠替代琼脂作为支持物,在培养 90 d后移栽;统计移栽 60 d后的成活率、萌芽数以及生根数.结果:铁皮石斛种子在N6 液体悬浮培养 30 d后,原球茎诱导率最高,可视化点播至 1/2 MS+5%马铃薯汁+15%香蕉汁+0.35 mg/L NAA+30 g/L蔗糖+7.5 g/L琼脂培养基,试管苗萌发密度均匀,不经过壮苗转接阶段即可长到 3~4 cm;转接至MS+7.5%香蕉汁+7.5%马铃薯汁+30 g/L蔗糖+2.0 mg/L 6-BA+0.5 mg/L NAA+3 cm装瓶高度椰糠∶珍珠岩=4∶1/瓶培养基后,生根状态良好,根系为绿色,不需要经过炼苗和洗苗环节,直接移栽成活率达到 96%以上.结论:采用液体悬浮培养铁皮石斛种子后,可视化播种,可以省去目前工艺的壮苗转接阶段,结合基质替代生根技术,可以降低种苗生产成本至少 30%,具有很好的应用推广价值.

Objective:To optimize the factory production process of Dendrobium officinale seedlings,in order to reduce the cost of seedling production and increase the survival rate of transplanting.Methods:Dendrobium officinale seeds were cultured in liquid suspen-sion for 30 days,and then 50 seeds were sucked up after the seeds turned into protocorms,visualized and sown into different formula-tions of medium,and the best medium was screened by the growth potential of test-tube seedlings after 120 days of cultivation;Coconut husk was used as a support instead of agar during the rooting stage,and the transplanting was carried out after 90 days of cultivation,the survival rate of transplanting,the number of sprouts,and the number of roots at 60 days after transplanting were statistically analyzed.Results:The highest induction rate of protocorms was observed after 30 days of N6 liquid suspension culture of Dendrobium officinale seeds.The germination density of test-tube seedlings was uniform after visualized and sown to 1/2 MS+5%potato juice+15%banana juice+0.35 mg/L NAA+30 g/L sucrose+7.5 g/L agar medium,and they could grow up to 3~4 cm without going through the stout transfer stage;The rooting status was good after transferring to MS+7.5%banana juice+7.5%banana juice+7.5%potato juice+30 g/L sucrose+2.0 mg/L 6-BA+0.5 mg/L NAA+3 cm bottled height coconut sugar:perlite 4∶1 per bottle,the rooting condition was good and the root system was green,and the survival rate of direct transplanting reached more than 96%without going through seedling refi-ning and seedling washing.Conclusion:The test results show that the use of seed liquid sprouting visualization precision sowing can e-liminate the strong seedling transfer stage of the current process,combined with the substrate alternative rooting technology,it can re-duce the seedling production cost of at least 30%or more,and has a good application and promotion value.
