目的:本研究旨在探究寒喘祖帕对过敏性哮喘(AAS)小鼠模型的抗炎药效学作用以及对肠道菌群结构的调节效应.方法:采用卵蛋白(OVA)致敏和激发的方法建立 AAS小鼠模型,分别给予地塞米松、寒喘祖帕进行灌胃给药干预,灌胃给药 3 周后使用动物肺功能仪检测各组小鼠的气道高反应性(AHR).收集小鼠血液标本及支气管肺泡灌洗液(BALF),进行细胞计数分析,采用 ELISA法检测BALF及血清中免疫球蛋白 E(IgE)、肿瘤坏死因子-α(TNF-α)、白介素(IL)-4、IL-5、IL-6、IL-13 的含量.取肺组织采用苏木精-伊红(HE)染色,观察小鼠肺组织病理变化.采用 16 S rRNA 测序技术分析小鼠肠道菌群的变化.结果:寒喘祖帕显著改善哮喘小鼠精神状态、活动度及体重,并延缓Penh值升高,降低 AHR.病理学分析表明,寒喘祖帕可减轻气管上皮损伤、肺间隔增厚及炎症浸润.肠道菌群分析显示,寒喘祖帕显著提高 Shannon和 Simpson 指数(P<0.05),恢复菌群α多样性.在门水平上,寒喘祖帕增加厚壁菌门、变形菌门等有益菌丰度,降低拟杆菌门比例;属水平上,普雷沃菌 Prevotellaceae_UCG_001、阿克曼菌属 Akkermansia等 9 个菌群被鉴定为关键生物标志物.结论:寒喘祖帕通过调节肠道菌群结构及多样性,缓解哮喘症状,其作用机制可能与菌群-免疫轴调控相关,为寒喘祖帕治疗哮喘提供了微生物组学依据.

Objective:This study aims to investigate the anti-inflammatory pharmacodynamic effects of Han-chuan Zupa on an allergic asthma(AAS)mouse model and its regulatory effects on the gut microbiota structure.Methods:An AAS mouse model was established using ovalbumin(OVA)sensitization and challenge.Mice were ad-ministered dexamethasone and hantazol via oral gavage.After three weeks of oral gavage,airway hyperresponsiveness(AHR)was assessed using an EMKA volume recorder.Blood samples and bronchoalveolar lavage fluid(BALF)were collected from the mice,and cell counts were analyzed.The levels of IgE,TNF-α,IL-4,IL-5,IL-6,and IL-13 in BALF and serum were detected using the ELISA method.Lung tissue was stained with hematoxylin and eosin(HE)to observe pathological changes in mouse lung tissue.The 16 S rRNA sequencing technique was used to analyze chan-ges in the intestinal microbiota of mice.Results:Hanchuan Zupa significantly improved the mental state,activity lev-el,and body weight of asthmatic mice(P<0.05),delayed the increase in Penh values,and reduced airway hyperre-sponsiveness(AHR).Pathological analysis indicated that Hanchuan Zupa alleviated tracheal epithelial damage,pul-monary interstitial thickening,and inflammatory infiltration.Gut microbiota analysis showed that Hanchuan Zupa significantly increased the Shannon and Simpson indices and restoredαdiversity of the microbiota.At the phylum level,Hanchuan Zupa increased the abundance of beneficial bacteria such as Firmicutes and Proteobacteria,while re-ducing the proportion of Bacteroidetes;at the genus level,nine bacterial communities,including Prevotellaceae_UCG_001 and Akkermansia,were identified as key biomarkers.Conclusion:Hanchuan Zupa alleviates asthma symptoms by regulating the structure and diversity of the gut microbiota.Its mechanism of action may be related to the regulation of the microbiota-immune axis,providing microbiological evidence for the use of Hanchuan Zupa in the treatment of asthm.
