基于内质网应激与自噬探讨炎症性肠病发病机制与中医药研究进展OA
Exploring the pathogenesis of inflammatory bowel disease based on endoplasmic reticulum stress and autophagy and the progress of traditional Chinese medicine intervention research
炎症性肠病(IBD)是一种慢性非特异性疾病,且发病率逐年上升,其发病机制尚未完全明确.目前研究结果表明,IBD的进展与内质网应激(ERS)、自噬密不可分.持续的 ERS可引起炎症反应、损伤肠黏膜屏障功能促引起进 IBD的发生,而自噬可以清除错误折叠蛋白的积累从而维持内质网稳态.中医药具有多靶点、多通路、副作用小等特点,在防治 IBD中具有显著临床疗效.本研究以 ERS与自噬为切入点,从炎症反应、肠黏膜屏障、免疫反应几方面探讨了其对 IBD发生发展的影响,并总结目前中药单体、复方及中医特色疗法通过调控 ERS及自噬治疗IBD的研究进展,为深入了解 IBD的发病机制提供理论支持,为中医药治疗 IBD提供新思路.
Inflammatory bowel disease(IBD)is a chronic non-specific disease and its incidence is increasing year by year,and its pathogenesis has not been fully clarified.The current findings suggest that the progression of IBD is inextricably linked to endoplasmic reticulum stress(ERS)and autophagy.Sustained ERS can cause inflamma-tory response and damage the barrier function of intestinal mucosa to promote the development of IBD,while autoph-agy can remove the accumulation of misfolded proteins to maintain endoplasmic reticulum homeostasis.With the characteristics of multi-targets,multi-pathways and low side-effects,TCM has significant clinical efficacy in the pre-vention and treatment of IBD.In this study,we took ERS and autophagy as the entry point,explored their effects on the development of IBD from the aspects of inflammatory response,intestinal mucosal barrier and immune response,and summarised the current progress of the research on the regulation of ERS and autophagy by traditional Chinese medicine(TCM)single-pharmaceuticals,compound formulas and TCM speciality therapies in the treatment of IBD,so as to provide theoretical support for the in-depth understanding of the pathogenesis of IBD,and to provide a new way of thinking for the treatment of IBD with TCM.
