海洋自絮凝菌在处理含盐污水中的应用研究OA

The application of Marine self-flocculating bacteria in the treatment of saline wastewater

通过微生物分离纯化技术从海洋底泥中成功筛选一株具有高效絮凝能力的菌株Y25.经革兰氏染色及16S rRNA序列分析鉴定,确定该菌株为革兰氏阳性菌,属于类芽孢杆菌属(Paenibacillus),与Paeniba-cillus chitinolyticus相似度最高.通过单因素实验系统考察温度、pH、溶解氧及盐度等环境参数对菌株生长特性的影响,并评估其在高盐废水处理中的潜在应用价值.实验结果表明:在达到24 h培养周期时,菌株Y25的絮凝活性达到峰值,其最适培养条件为盐度3%(w/v)、温度30℃、pH 7及转速200 r/min;在模拟含盐污水体系中,该菌对化学需氧量(COD)、总磷(TP)和氨氮(NH4+-N)的去除率分别可达89.1%、76.7%和88.6%.证明了自絮凝菌株在含盐污水处理方面的应用价值.

The microbial isolation and purification technique was used to successfully screen a strain Y25 with high flocculation activity from marine mud,which was identified as a Gram-positive bacterium be-longing to the genus Paenibacillus,with the highest degree of similarity to Paenibacillus chitinolyticus,by Gram staining and 16S rRNA sequence analysis.The effects of environmental parameters such as tem-perature,pH,dissolved oxygen and salinity on the growth characteristics of the strains were investigated by a one-factor experimental system,and their potential application value in the treatment of high-salt wastewater was evaluated.The experimental results showed that the flocculation activity of strain Y25 peaked at 26 h of incubation cycle,and its optimal culture conditions were 3%salinity(w/v),30℃,pH 7.0,and 200 r/min;in the simulated saline wastewater system,the removal of chemical oxygen demand(COD),total phosphorus(TP),and ammonia nitrogen(NH4+-N)by this strain could be up to 89.1%,76.7%,and 88.6%,respectively.The self-flocculating strains proved to be valuable for the application in the treatment of saline wastewater.
