不同氮源浓度配比对苹果砧木G935试管苗生长及扩繁的影响研究OA
The Impact of Different Nitrogen Source Concentration Ratios on the Growth and Propagation of Apple Rootstock G935 in Vitro
[目的]选用苹果砧木G935试管苗,通过调整MS培养基中KNO3和NH4NO3的浓度配比,筛选出最有利于G935试管苗高效扩繁与优质生长的氮源水平,为建立该砧木的高效快繁体系提供理论支撑和配方优化方案.[方法]以苹果砧木G935的试管苗为材料,选取带2.0 cm叶片的茎段作为外植体,以MS培养基为基础,设置5个不同浓度KNO3和NH4NO3的处理,对照CK(标准MS氮源:KNO3 1.9 g/L,NH4NO3 1.65 g/L),培养5周后,测定外植体的5个生长指标(扩繁系数、株高、节间长度、茎粗、叶绿素),并进行主成分分析.[结果]适度降低氮源总量及其比例可显著影响G935试管苗的生长表现,其中处理T2(KNO3 1.3 g/L+NH4NO3 1.1 g/L)在综合指标评价中表现最优,尤其在扩繁系数、节间长度控制及叶绿素含量方面达到最佳平衡.主成分分析结果进一步确认了处理T2的综合评价值最高.[结论]研究结果为苹果砧木高效离体快繁体系的氮营养优化提供了理论依据与实践参考.
[Objective]Apple rootstock G935 seedlings in vitro were selected,and the concentration ratios of KNO3 and NH4NO3 in MS medium were adjusted to screen for the nitrogen source levels most conducive to the efficient propagation and high-quality growth of G935 test-tube seedlings.This provides theoretical support and formula optimization for establishing an efficient rapid propagation system for this rootstock.[Methods]Test-tube seedlings of apple rootstock G935 were used as the material,with stem segments bearing 2.0 cm leaves as the explants.Based on MS medium,five different concentration treat-ments of KNO3 and NH4NO3 were designed,with the control CK(standard MS nitrogen source:KNO3 1.9 g/L,NH4NO3 1.65 g/L).After five weeks of culture,five growth indices of the explants(propagation coefficient,plant height,internode length,stem diameter,chlorophyll)were measured and subjected to principal component analysis.[Results]Moderate reduc-tion in the total amount and proportion of nitrogen sources significantly affected the growth performance of G935 test-tube seedlings.Among them,treatment T2(KNO3 1.3 g/L+NH4NO3 1.1 g/L)showed the best performance in the compre-hensive index evaluation,especially in achieving the optimal balance in the propagation coefficient,internode length control,and chlorophyll content.The results of principal component analysis further confirmed that treatment T2 had the highest com-prehensive evaluation value.[Conclusion]A theoretical basis and practical reference are provided for the optimization of nitro-gen nutrition in the efficient in vitro rapid propagation system of apple rootstock.
