漆树子代遗传多样性分析与指纹图谱构建OA
Analysis of genetic diversity and fingerprinting for offspring of Toxicodendron vernicifluum
为探明云南省漆树家系子代的遗传多样性,促进漆树特异品种的选育及应用,在云南省昭通市、怒江州、迪庆州采集 36 个漆树家系的 36 份种子,对其进行催芽、播种、育苗,获得子代嫩叶材料后提取基因组DNA,用简单序列重复-聚合酶链式反应(SSR-PCR)扩增,再利用毛细管电泳和SSR技术对 36 份漆树子代种质材料进行位点检测,最终进行遗传多样性评估,并利用SSR分子标记建立家系DNA指纹图谱.PCR分析结果表明,9 个位点共检测到 26.667 个观测等位基因,平均观测等位基因数为 2.963,有效等位基因数平均值为 2.280,Shannon's信息指数平均值为 0.835,观测杂合度平均值为 0.527,期望杂合度平均值为 0.484,多态信息含量平均值为 0.534,基因流平均值为 2.131.36份漆树子代材料整体上表现出较高的遗传相似度,聚类分析将供试材料分为 3 个类群.漆树家系子代具有较为丰富的遗传多样性,群体间产生很小的遗传分化,存在较为复杂的遗传背景.多位点匹配分析确定了 c26126、M66、M156 和Tox003 为核心引物,利用这4 对核心引物可构建 36 份漆树子代材料的指纹图谱.
This study aims to investigate the genetic diversity of offspring of Toxicodendron vernicifluum families in Yunnan Province and promote the breeding and application of unique T.vernicifluum varieties.Thirty-six seed samples from 36 T.vernicifluum families were collected in Zhaotong City,Nujiang Prefecture,and Diqing Prefecture of Yunnan Province.The seeds were treated for germination and then sown,and tender leaves of the seedlings were collected for genomic DNA extraction.Simple sequence repeat-polymerase chain reaction(SSR-PCR)was used for amplification,followed by capillary electrophoresis and SSR for locus detection of the 36 T.vernicifluum offspring families.Genetic diversity was subsequently assessed,and SSRs were used to establish DNA fingerprints of the families.PCR revealed 26.667 observed alleles across 9 loci,with an average number of observed alleles being 2.963.The average number of effective alleles,average Shannon's information index,average observed heterozygosity,average expected heterozygosity,average polymorphic information content,and average gene flow were 2.280,0.835,0.527,0.484,0.534,and 2.131,respectively.Overall,the 36 T.vernicifluum offspring families showed high genetic similarity,and cluster analysis classified them into 3 groups.The offspring families of T.vernicifluum exhibited rich genetic diversity,with minimal genetic differentiation among populations and a relatively complex genetic background.Multi-locus matching analysis identified c26126,M66,M156,and Tox003 as core primers,which could be used to construct DNA fingerprints for the 36 T.vernicifluum offspring families.
