目的:探究超短波对急性肺损伤大鼠炎性反应及肺泡上皮细胞凋亡的影响.方法:将30只雄性SD大鼠随机分为对照组、LPS损伤组、超短波组,每组10只.通过脂多糖尾静脉注射建立急性肺损伤模型,超短波组在造模后即刻、24h、48h予超短波干预15 min;对照组和LPS损伤组置于不通电的超短波电极板中间,干预结束后24h处死实验大鼠.ELISA检测血清中IL-1β、IL-10的表达,HE染色评估肺损伤程度,TUNEL法检测肺泡上皮细胞凋亡变化,PCR及Western-blot分别检测caspase-3、caspase-8 mRNA和蛋白表达.结果:与对照组相比,LPS损伤组大鼠血清IL-1β表达显著升高、IL-10表达显著降低(P<0.01),肺组织结构显著受损,肺损伤评分和肺泡上皮细胞凋亡率显著升高(P<0.01),caspase-3、caspase-8 mRNA和蛋白表达显著升高(P<0.01).经超短波干预后,LPS损伤大鼠血清IL-1β表达显著降低、IL-10表达显著升高(P<0.01),肺损伤评分及肺泡上皮细胞凋亡率显著下降(P<0.01),caspase-3、caspase-8 mRNA和蛋白表达显著下降(P<0.01).结论:超短波疗法通过下调caspase-8/caspase-3/IL-1β信号通路,抑制肺泡上皮细胞凋亡,减轻急性肺损伤炎性反应.

Objective:To investigate the effects of ultrashort waves on anti-inflammatory capacity and cell apoptosis in LPS-induced acute lung injury rats.Methods:Thirty male SD rats were randomly divided into the control group,the LPS injury group and the ultrashort wave group,with 10 rats in each group.The acute lung injury model was established by lipopolysaccharide tail vein injection,and the ultrashort wave group was given ultrashort wave intervention for 15 min immediately,24 h and 48 h after modelling;the control group and the LPS injury group were placed in the middle of the unelectrified ultrashort wave electrode plate.Immediately after the intervention,the rats were put to death,the expression of IL-1β and IL-10 in serum was detected by ELISA,the degree of lung injury was assessed by HE staining,the apoptotic changes of alveolar epithelial cells were detected by TUNEL,and the expression of caspase-3 and caspase-8 mRNA and protein were detected by PCR and Western-blot,respectively.Results:Compared with the control group,serum IL-1β expression was significantly higher and IL-10 expression was significantly lower in the LPS-injured group(P<0.01),lung tissue structure was significantly damaged,lung injury score and apoptosis rate of alveolar epithelial cells were significantly higher(P<0.01),and the expression of caspase-3 and caspase-8 mRNA and protein was significantly higher(P<0.01).After ultrashort wave intervention,LPS-injured rats showed a significant decrease in serum IL-1β expression,a significant increase in IL-10 expression(P<0.01),a significant decrease in lung injury score and apoptosis rate of alveolar epithelial cells(P<0.01),and a significant decrease in caspase-3 and caspase-8 mRNA and protein expression(P<0.01).Conclusion:Ultrashort wave therapy exerts anti-inflammatory effects on lipopolysaccharide-induced acute lung injury by inhibiting alveolar epithelial cell apoptosis by downregulating the caspase-8/caspase-3/IL-1β signaling pathway.
