多重荧光PCR法鉴别半夏药材及饮片中的掺混伪品OA北大核心
Development and Application of Multiplex Fluorescent PCR for Identification of Pinellia ternate from its Counterfeits
目的:建立多重荧光PCR法鉴别半夏以及常见混伪品虎掌、水半夏、天南星的方法,并验证其可行性,为半夏质量标准的制定提供方法学支持.方法:对多重荧光PCR扩增体系进行优化,同时对该方法的特异性、灵敏度、检出限进行考察,并对前期性状鉴定疑似为半夏伪品的 15 批药材及饮片进行实测验证.结果:研究建立的方法检测出 8 批为虎掌、3 批为水半夏、4 批为非半夏其他混淆品,所测结果与测序结果完全一致.该方法在同科属常见近缘物种中无交叉反应,仅需要皮克(pg)级的DNA模板量即可检测,并且可以检出掺伪率仅为 5%的混伪品.结论:该研究建立的多重荧光PCR法具有灵敏、快速、准确等优势,可以作为半夏药材及饮片中掺混伪品的特异性检测方法推广使用.
Objective:To establish a multiplex fluorescent PCR method for identification of Pinellia ternate and its common counter-feits Pinellia pedatisecta,Typhonium flagelliforme,Arisaema erubescens,and to verify its feasibility,so as to provide methodological support for formulating the quality standard of Pinellia ternate.Methods:The multiplex PCR system was optimized,and the specificity,sensitivity,detection limit of the method were investigated.The 15 batches of medicinal materials and decoction pieces suspeeted to be Pinellia ternate counterfeit in the earlier stage were tested and verified.Results:The method established in this study detected 8 batches of Pinellia pedati-secta,3 batches of Typhonium flagelliforme and 4 batches of non-Pinellia ternate other confounding products,and the measured results were completely consistent with the sequencing results.The method had no cross-reaction in the common related species of the same family,and only needed the template amount of pg DNA to detect,and could detect the mixed counterfeits with only 5%adulteration rate.Con-clusion:The established multiplex fluorescent PCR method has the advantages of sensitivity,speed and accuracy,and can be widely used as a specific detection method for Pinellia ternate medicinal materials and decoction pieces and its common counterfeits.
