Quality Evaluation of Taxilli Herba Based on HPLC Fingerprint and Multi-component Quantitative Analysis

目的:建立桑寄生HPLC指纹图谱及没食子酸、萹蓄苷、槲皮苷、儿茶素、异槲皮苷、表儿茶素6 个成分含测定方法,并对不同产地的桑寄生质量进行评价.方法:采用GL Sciences InertSustain AQ-C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈-0.1%磷酸溶液为流动相,梯度洗脱;检测波长为 230 nm;柱温为 30℃;流速为 1.0 mL/min;进样量为 10 μL.构建桑寄生HPLC指纹图谱,同时测定其中 6 个成分的含量.结合聚类分析(CA)、主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)和熵权-TOPSIS分析进行质量评价.结果:建立的桑寄生HPLC指纹图谱标定了 14 个共有峰;证实 5 个为引起质量差异的标志物;熵权-TOPSIS法分析结果显示广东、广西产地的桑寄生质量较佳;同时发现不同产地桑寄生药材中没食子酸、儿茶素、表儿茶素、异槲皮苷、萹蓄苷、槲皮苷含量差异明显.结论:建立的桑寄生HPLC指纹图谱及含量测定方法合理可靠,化学计量学结合熵权-TOPSIS法可用于评价桑寄生的质量,为其质量控制提供依据.

Objective:To establish the HPLC fingerprint of Taxilli Herba and the determination methods of gallic acid,avicularin,quercitrin,catechin,isoquercitrin and epicatechin,to evaluate the quality of Taxilli Herba from different habitats.Methods:The chroma-tography was performed on the GL Sciences InertSustain AQ-C18(250 mm×4.6 mm,5 μm)chromatographic column,the mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution with gradient elution,detection wavelength was 230 nm,column temperature was 30℃,flow rate was 1.0 mL/min,and injection volume was 10 μL.The HPLC fingerprint of Taxilli Herba was constructed,and the con-tents of six components were simultaneously determined.A combinations of cluster analysis(CA),principal component analysis(PCA),orthogonal partial least squares discriminant analysis(OPLS-DA)and entropy weight-TOPSIS analysis was used to evaluate the quality.Results:HPLC fingerprint of Taxilli Herba was established and 14 common peaks were calibrated.5 markers of quality difference were confirmed.The results of entropy weight-TOPSIS analysis showed that the quality of Taxilli Herba from Guangdong,Guangxi was better.The contents of gallic acid,catechin,epicatechin,isoquercetin,avicularin and quercetin were significantly different in different areas.Conclusion:The HPLC fingerprint and content determination method of Taxilli Herba is reasonable and reliable.Chemometrics combined with entropy weight-TOPSIS method can be used to evaluate the quality of Taxilli Herba and provide basis for its quality control.
