野八角果实的化学成分研究OA北大核心
Chemical Constituents from the Fruits of Illicium simonsii
目的:研究野八角Illicium simonsii Maxim.果实的化学成分.方法:采用硅胶柱、Sephadex LH-20 葡聚糖凝胶柱、RP-C18 柱以及半制备高效液相等色谱技术对野八角果实醇提物的乙酸乙酯部位进行分离纯化,并利用波谱数据结合文献鉴定化合物结构.采用MTT法测试化合物的体外抗人结肠癌细胞(HCT-116)活性.结果:从野八角中分离得到 18 个化合物,分别鉴别为:henrylactone A(1)、oligandrumin D(2)、veranisatin D(3)、布卢门醇A(4)、布卢门醇B(5)、青刺尖木脂醇(6)、7S,8R-二氢去氢双松柏醇(7)、sinensiol G(8)、去氢双松柏醇(9)、rhoifoline B(10)、4-methoxy-N-methyl-2-quinolone(11)、N-反式-对香豆酰基酪胺(12)、对羟基苯甲醛(13)、香草醇(14)、槲皮素(15)、mexoticin(16)、samwirin(17)、左旋马尾松树脂醇(18).体外试验研究发现化合物 1、11 对 HCT-116 细胞具有抑制作用.结论:其中,化合物 4~6、8、10~12、14、16、17 为首次从该属植物中分离得到,化合物 18 为首次从该植物中分离得到.化合物 1、11 表现出潜在的抗肿瘤活性,IC50 分别为(3.18±0.11)、(2.43±0.12)μmol/L.
Objective:To study the chemical constituents from the fruits of Illicium simonsii.Methods:The compounds from the eth-yl acetate fraction of the ethanol extract of Illicium simonsii fruits were isolated and purified by silica gel column chromatography,Sepha-dex LH-20 gel column chromatography,RP-C18 column chromatography and semi-preparative high performance liquid chromatography,and the structures of the compounds were identified by spectral data and literature.The in vitro anti-human colon cancer cell(HCT-116)activity of the compounds was tested by MTT assay.Results:Eighteen compounds were isolated from the fruits of Illicium simonsii and i-dentified as henrylactone A(1),oligandrumin D(2),veranisatin D(3),blumenol A(4),blumenol B(5),prinsepiol(6),7S,8R-di-hydrodehydrodiconiferyl alcohol(7),sinensiol G(8),dehydrodiconiferyl alcohol(9),rhoifoline B(10),4-methoxy-N-methyl-2-quinolo-ne(11),trans-N-p-coumaroyltyramine(12),p-hydroxybenzaldehyde(13),vanillyl alcohol(14),quercetin(15),mexoticin(16),samwi-rin(17),(-)-massoniresinol(18),respectively.In vitro experiments showed that compounds 1 and 11 had inhibitory effect on HCT-116 cells.Conclusion:Among them,compounds 4 to 6,8,10 to 12,14,16,17 are isolated from Illicium genus for the first time,compound 18 is isolated from Illicium simonsii for the first time.Compounds 1 and 11 show potential antitumor activity with IC50 of(3.18±0.11)μmol/L and(2.43±0.12)μmol/L,respectively.
