目的:开发一种基于香豆素并吡啶结构的pH敏感荧光染料(Coumarin-pyridine,Cou-py),用于碱性pH值响应,并实现细胞成像及试纸检测.方法:以4-甲基-7-二乙胺基香豆素为原料,先后与Vilsmeier-Haack试剂和氨水进行反应,得到Cou-py,并分别在溶液、试纸和细胞中探究其对碱性pH值的响应性能.结果:成功合成Cou-py,吸收和荧光光谱分析表明,在碱性环境中Cou-py的吸收峰由405 nm蓝移至365 nm,可肉眼观察到溶液由黄色转变为无色,并且随着pH值升高,465 nm处的荧光发射逐渐增强,在pH 10.5时达到pH 6.5时的28倍(线性响应范围pH 7.0~10.0,R2=0.995).细胞实验表明,Cou-py具有低细胞毒性和良好的光稳定性,成功可视化细胞内碱性环境.试纸应用显示,碱性条件触发Cou-py蓝色荧光"开启",实现快速可视化检测.结论:提出了一种pH敏感荧光染料即Cou-py,不仅在溶液体系中表现出比色和荧光的双重响应,还能够对细胞内的碱性环境进行成像.该染料可负载在试纸上,通过蓝色荧光"开启"实现对碱性pH值的快速和便捷的检测.本研究为细胞内碱性环境监测提供了一种高效的工具.

Objective:We developed a pH-sensitive fluorescent dye based on the coumarin-pyridine(Cou-py)structure to detect alkaline pH,which enables cell imaging and test strip detection.Methods:Cou-py was synthesized by sequentially reacting 4-methyl-7-diethylaminocoumarin with the Vilsmeier-Haack reagent and aqueous ammonia.Its pH-responsive performance was systematically investigated in solution,on test strip,and in cellular environment under alkaline conditions.Results:Cou-py was successfully synthesized,and absorption and fluorescence spectroscopic analyses demonstrated that its absorption peak exhibited a blue shift from 405 nm to 365 nm under alkaline conditions.The solution could be observed from yellow to colorless by naked eyes,accompanied by a gradual enhancement of fluorescence emission at 465 nm with increasing pH,showing a 28-fold enhancement at pH 10.5 versus pH 6.5(linear range:pH 7.0~10.0,R2=0.995).Cell experiments demonstrated that Cou-py exhibited low cytotoxicity and excellent photostability,successfully visualizing the alkaline cellular environment.Test strip application demonstrated that Cou-py displayed a blue fluorescence"turn-on"response triggered by alkaline conditions,achieving rapid visual detection.Conclusion:A pH-sensitive fluorescent dye,Cou-py,was proposed which not only exhibited a dual colorimetric and fluorescent response under alkaline conditions in solution systems,but also was capable of imaging the alkaline cellular environment.This dye could be loaded onto test strip for quick and convenient detection of alkaline pH through blue fluorescence"turn-on".This study provides an efficient tool for monitoring intracellular alkaline environments.
