藻类色素提取方式的比较研究OA北大核心
A comparison of varied extraction methods for algal pigments
藻类色素通过吸收和传递光能参与光合作用,在不同藻类中色素组成和含量状况存在明显差异,因此,色素分析是进行藻类光合生理研究和藻种辅助鉴定的重要手段,在浮游植物类群组成和生物量评估等生态学研究中也有广泛应用.针对藻类色素分析,目前已经建立了多种色素提取和分析方法.本研究对比分析了 5 种细胞破碎色素提取方式(超声处理、手动研磨、冷冻研磨、手动研磨+超声处理、冷冻研磨+超声处理)对微藻和大型藻中色素提取的效果,结果表明:(1)超声处理对具有坚硬硅质细胞壁的硅藻色素提取效果更好,而研磨或超声处理对不具细胞壁的甲藻色素提取效果相似;(2)5 种提取方式对 2 种大型藻中色素的提取效果存在较大差异,在铜藻(Sargassum horneri)色素的提取中尤为明显.对浒苔(Ulva prolifera)中色素的提取效果手动研磨优于冷冻研磨,而冷冻研磨对铜藻色素的提取效果更好,若辅以超声处理可以更好地促进铜藻色素提取;(3)综合来看,冷冻研磨+超声处理对于微藻和大型藻样品中色素提取的效果最好.该提取方式操作简便、高效省时,在今后藻类色素分析工作中应进一步推广应用.
Algal pigments participate in photosynthesis by absorbing and transferring light energy.The composition and content of pigments vary among algae.Therefore,pigment analysis is crucial for studying photosynthetic physiology and identifying algal taxa.It has been widely applied in ecological explorations of the phytoplankton community composition and biomass assessment.Many extraction and analytical methods for algal pigments have been developed.This study compared five cell disruption and pigment extraction methods—sonication,manual grinding,freeze grinding,manual grinding combined with sonication,and freeze grinding combined with soni-cation—in micro-and macro-algae.The major findings were:(1)Sonication was particularly effective in extracting pigments from diatoms with rigid silica cell walls,unlike grinding and sonication in dinoflagellates without cell walls.(2)The five methods had notable variations in pigment extraction efficiency from two species of macroalgae,especially the brown alga Sargassum horneri.Manual grinding was more effective than freeze-grinding in the green alga Ulva prolifera,unlike in S.horneri.Sonication could further improve elicitation from S.horneri.(3)Among the five methods,freeze grinding combined with sonication was most effective in pigment extraction from micro-and macro-algal samples.Such a protocol is easy to operate,efficient,and time-saving.It presents wide applicabil-ity in future studies.
