药用蜘蛛棒毛络新妇及其混淆品的分子鉴定OA北大核心

目的:由于蜘蛛类的物种繁多,常导致该类药材相互混淆,存在用药的安全隐患,亟待建立一种能准确识别棒毛络新妇及其混淆品的分子鉴定方法.方法:针对采集的 70 份样品提取DNA,分析其COⅠ序列的特征、计算种内和种间遗传距离并构建ML系统发育树,评估COⅠ序列作为鉴定棒毛络新妇及其混淆品的有效性.使用R语言绘图软件将分子条形码转换成QR二维码,建立专属的DNA条形码库,并对市售的 8 份蜘蛛类药材进行实测验证.结果:棒毛络新妇的COⅠ序列长度为 603 bp,GC含量为 31.8%~32.5%,存在 16 个碱基变异位点,共有 14种单倍型(Hd=0.8236),表明样本群体具有丰富的多样性.种间平均遗传距离(0.1685)显著大于种内平均遗传距离(0.0034),且与DNA Barcoding Gap检验结果一致,说明COⅠ序列与其他物种之间具有一定的DNA条形码间隙.在构建的系统发育树中,不同的物种均各自聚为单支.实测验证的 8 份市售样品中有 6 份样品鉴定为棒毛络新妇,2 份伪品也被准确识别,其准确率达 100%.结论:该研究建立的COⅠ序列分子鉴定方法专属性强,可以有效地解决棒毛络新妇完整药材及其粉末的物种鉴定问题.

Objective:To establish a molecular identification method that can accurately identify Trichonephila clavata and its con-fused products to solve the confusion of medicinal materials and the hidden danger of drug use caused by the diversity of spider species.Methods:DNA was extracted from 70 samples,COⅠ sequence characteristics were analyzed,intraspecious and interspecific genetic dis-tances were calculated,and ML phylogenetic trees were constructed to evaluate the validity of COⅠ sequence as an identification meth-od of Trichonephila clavata and its confounding products.R language mapping software was used to convert molecular barcodes into QR codes,establish a dedicated DNA barcode library,and verify 8 commercially available spiders.Results:The length of COⅠ sequence of Trichonephila clavata was 603 bp,the content of GC was 31.8%to 32.5%,there were 16 base variation sites,and there were 14 haplo-types(Hd=0.8236),indicating that the sample population had rich diversity.The average interspecific genetic distance(0.1685)was significantly greater than the average intrasecific genetic distance(0.0034),which was consistent with the DNA Barcoding Gap test re-sults,indicating that there was a certain DNA barcode gap between COⅠ sequence and other species.In the constructed phylogenetic tree,different species were grouped into single branches.Among the 8 commercial samples verified by actual measurement,6 samples were identified as Trichonephila clavata,and 2 counterfeits were also accurately identified,with an accuracy of 100%.Conclusion:The molecular identification method of COⅠ sequence established in this study is highly specific,and can effectively solve the species iden-tification problem of the complete herbal medicine and its powder of Trichonephila clavata.
