目的:比较不同年限茅苍术药材的性状特征、主要初生代谢物、挥发油及其主要活性成分的分布差异,为阐明茅苍术有效成分的分布特点和积累规律提供科学依据.方法:利用形态学直观和量化的方法进行比较,分析不同年限药材断面、质地和油室密度;分别采用考马斯亮蓝染色法、蒽醌比色法和水蒸气蒸馏法测定可溶性蛋白、可溶性糖和淀粉以及总挥发油的含量;使用高效液相色谱法(HPLC)和气质联用(GC-MS)法测定苍术素、苍术酮等特征性成分含量.结果:随着年份增加药材断面颜色加深,质地更加坚硬;断面中油室变大,颜色加深,油室面积与总面积之比逐年增大;鲜药材密度随着年份的增加而减小,干药材密度随着年份的增加逐渐增大;可溶性蛋白和可溶性糖的含量均随着年份增加而减少,淀粉含量随着年份增加逐渐增多.随着年份的增加,药材中总挥发油和苍术素的含量逐渐升高,苍术酮、β-桉叶醇的含量逐渐降低,而茅术醇含量无显著差异.结论:不同年限茅苍术药材中主要初生代谢物和次生代谢物空间分布及其含量存在显著差异,该研究结果为进一步探讨生长年限对茅苍术药材品质的影响机制提供了理论支撑.

Objective:To compare the distributional difference of characteristics,main primary metabolites,volatile oil and main active components of Atractylodes lancea in different years,so as to provide scientific basis for elucidating the distribution characteristics and accumulation rules of active components of Atractylodes lancea.Methods:The section,texture and oil chamber density of herbs in different years were andyzed by visual and quantitative morphological methods.The soluble protein,soluble sugar and starch,total vola-tile oil contents were determined by Coomassie bright blue staining,anthraquinone colorimetric,steam distillation methods respectively.The contents of characteristic components such as atractylodin and atractylon were determined by HPLC and GC-MS.Results:With the increase of years,the color of the section was deepened,the texture was harder,the oil chamber in the section was enlarged,color was deepened,the ratio of oil chamber area to total area increased year by year.The density of fresh medicinal materials decreased with the increase of years,and the density of dry medicinal materials increased with the increase of years.The contents of soluble protein and sol-uble sugar decreased with the increase of years,and the contents of starch gradually increased with the increase of years.With the in-crease of years,the total volatile oil and atractylodin contents in the medicinal materials gradually increased,while the contents of atrac-tylon and β-eucalyptol gradually decreased.However,there was no significant difference in the content of hinesol.Conclusion:There are significant differences in the spatial distribution and content of primary and secondary metabolites in different age groups.The results provide theoretical support for further study on the mechanism of the influence of growth age on the quality of Atractylodes lancea.
