一贯煎影响肝癌干细胞抑制肝细胞癌的作用机制研究OA
Mechanistic study of Yiguanjian Decoction in suppressing hepatocellular carcinoma by affecting liver cancer stem cells
目的 探讨一贯煎(YGJ)及其成分通过赖氨酰氧化酶样蛋白 2(LOXL2)影响肝癌干细胞(LCSCs)调控肝细胞癌(HCC)的作用机制.方法 分选CD133+LCSCs并构建沉默LOXL2 的LCSCs,进行细胞实验.YGJ干预沉默LOXL2 的LCSCs.经网络药理学筛选YGJ主要活性成分是槲皮素和异补骨脂黄酮,异补骨脂黄酮与槲皮素均属二氢黄酮类且二者结构相似.已明确异补骨脂黄酮对HCC增殖具有抑制作用.槲皮素经CCK8 法检测抑制LCSCs增殖的情况.构建LOXL2 及其下游因子非受体酪氨酸蛋白激酶SRC、磷脂酰肌醇 4,5-二磷酸 3 激酶催化α亚型(PIK3CA)过表达质粒转染LCSCs,双萤光素酶报告实验检测槲皮素对LOXL2 及SRC、PIK3CA的调控.建立荷人LCSCs裸鼠皮下移植瘤模型,并用药干预后验证各组瘤体组织中LOXL2、SRC、PIK3CA对HCC的影响.结果 沉默LOXL2 能抑制LCSCs的增殖、侵袭及迁移.YGJ影响LOXL2 表达抑制LCSCs增殖并促进其凋亡.槲皮素通过下调LOXL2 及下游因子SRC、PIK3CA基因的表达抑制LCSCs增殖,进一步抑制裸鼠体内LCSCs种植瘤生长.结论 YGJ及槲皮素通过下调LOXL2 及其下游SRC、PIK3CA基因影响LCSCs,进一步抑制HCC进展.
Objective To investigate the mechanism of action of Yiguanjian Decoction(YGJ)and its constituents in regulating hepatocellular carcinoma(HCC)by affecting liver cancer stem cells(LCSCs)through lysyl-oxidase like-2(LOXL2).Methods CD133+LCSCs were sorted and constructed to down-regulate LOXL2,cellular experiments were performed,and YGJ intervened to silence LOXL2 LCSCs.Through network pharmacology screening,the primary active components of YGJ were identified as quercetin and isoflavone.Both quercetin and isoflavone belong to the dihydroflavone class and share structural similarities.It has been established that isoflavone inhibits the proliferation of HCC.Inhibition of LCSC Proliferation by Quercetin as Detected by the CCK-8 Assay.Construction of LOXL2 and its downstream factors proto-oncogene tyrosine-protein kinase SRC,phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit alpha isoform(PIK3CA)LCSCs were transfected with the overexpression plasmid of PIK3CA.Dual luciferase reporter assay was performed to detect the regulation of LOXL2 and SRC,PIK3CA by quercetin.Establishment of subcutaneous grafted tumor model in nude mice with LCSCs and validation of the effects of LOXL2,SRC,and PIK3CA on HCC in tumor tissues of nude mice in each group after drug intervention.Results Knockdown of LOXL2 inhibited the proliferation,invasion,and migration of LCSCs.YGJ affected LOXL2 expression to inhibit the proliferation and promote apoptosis of LCSCs.Quercetin inhibited the proliferation of LCSCs by down-regulating the expression of LOXL2 and downstream factors SRC and PIK3CA genes,and further inhibited the growth of LCSCs implantation tumors in nude mice in vivo.Conclusion YGJ and quercetin affect LCSCs by down-regulating LOXL2 and its downstream SRC and PIK3CA genes to further inhibit HCC progression.
