目的 研究刺猬皮的氨基酸、微量元素成分,并初步分析其提取物的抗炎作用.方法 采用超高效液相色谱-串联质谱法分析刺猬皮中氨基酸种类及总含量;采用火焰原子吸收光谱法检测刺猬皮中各微量元素的含量;建立急性炎症和慢性炎症实验模型,观察刺猬皮提取物的抗炎效果,测定足匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)和髓过氧化物酶(MPO)水平,苏木精-伊红(HE)染色观察足组织病理学变化.结果 刺猬皮中共含有 17 种氨基酸,并含有镁、铁等微量元素;刺猬皮提取物各剂量组中耳廓肿胀度、足肿胀度、棉球肉芽肿干重、腹腔毛细血管通透性、足组织MDA和MPO水平、中性粒细胞浸润均低于模型组,SOD水平高于模型组(P<0.05 或P<0.01).结论 刺猬皮提取物能够有效抑制小鼠炎症反应,并可减轻机体氧化应激水平,该作用可能与刺猬皮中含有多种具有抗炎作用的氨基酸、微量元素有关.

Objective To analyze the amino acids and trace elements in hedgehog skin and preliminarily investigate the anti-inflammatory effects of its extracts.Methods Ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)was employed to determine the types and total content of amino acids.Flame atomic absorption spectrometry was used to detect trace elements.Acute and chronic inflammatory models were established to evaluate the anti-inflammatory effects of hedgehog skin extracts.Levels of superoxide dismutase(SOD),malondialdehyde(MDA),and myeloperoxidase(MPO)in foot homogenates were measured.Pathological changes in foot tissues were observed via hematoxylin-eosin(HE)staining.Results Hedgehog skin contained 17 amino acids and trace elements such as magnesium(Mg)and iron(Fe).Compared with the model group,all dose groups of hedgehog skin extracts showed reduced auricular swelling,foot swelling,dry weight of cotton ball granuloma,peritoneal capillary permeability,MDA and MPO levels,and neutrophil infiltration,while SOD levels were elevated(P<0.05 or P<0.01).Conclusion Hedgehog skin extract can effectively inhibit the inflammatory response of mice and reduce the level of oxidative stress in the body,which may be related to the presence of a variety of amino acids and trace elements with anti-inflammatory effects in hedgehog skin.
