镁离子体外促进巨噬细胞极化的作用研究OA
Study on the effect of magnesium ions on the polarization of macrophages in vitro
目的 探讨不同浓度的镁离子对巨噬细胞增殖及极化的影响.方法 使用含有不同镁离子浓度的培养基培养巨噬细胞,通过细胞计数试剂盒(CCK8)检测各组巨噬细胞的增殖情况,流式细胞术检测各组巨噬细胞的极化状态,实时荧光定量聚合酶链式反应(RT-qPCR)评估各组巨噬细胞极化相关基因表达水平.结果 50、100、200 μg/ml的氯化镁对巨噬细胞有明显促增殖作用(P<0.05),300、400、500 μg/ml氯化镁对巨噬细胞的增殖分化有明显抑制作用(P<0.05).同时,50、100、200 μg/ml的氯化镁能够明显促进巨噬细胞向M2型极化以及促进极化相关基因表达(P<0.05),300、400 μg/ml氯化镁促进巨噬细胞向M1型极化(P<0.05).结论 50、100、200 μg/ml的氯化镁能够明显促进巨噬细胞增殖分化以及向M2 型极化,300、400 μg/ml的氯化镁能够抑制巨噬细胞增殖分化以及促进巨噬细胞向M1 型极化.
Objective To investigate the effects of different concentrations of magnesium ions on the proliferation and polarization of macrophages.Methods Macrophages were cultured in media with different concentrations of magnesium ions.The proliferation of macrophages in each group was detected by cell counting kit-8(CCK8),and the polarization state of macrophages in each group was detected by flow cytometry.The expression levels of polarization-related genes in macrophages of each group were evaluated by real-time quantitative polymerase chain reaction(RT-qPCR).Results MgCl2 at 50,100,and 200 μg/ml had a significant pro-proliferative effect on macrophages(P<0.05),and MgCl2 at 300,400,and 500 μg/ml had a significant inhibitory effect on the proliferation and differentiation of macrophages(P<0.05).Meanwhile,50,100,and 200 μg/ml MgCl2 could significantly promote the polarization of macrophages to M2 type as well as promote the expression of polarization-related genes(P<0.05),and 300 and 400 μg/ml MgCl2 promoted the polarization of macrophages to M1 type(P<0.05).Conclusion 50,100,and 200μg/ml MgCl2 could significantly promote the proliferation and differentiation of macrophages and their polarization to M2 type,while 300 and 400 μg/ml MgCl2 could inhibit the proliferation and differentiation of macrophages and promote their polarization to M1 type.
