现场快速筛查即食海参中副溶血弧菌污染的方法OA
A Simple and Rapid Method for the Screening of Vibrio parahaemolyticus Contamination in Ready-to-Eat Sea Cucumber On-site
基于电子微生物生长分析仪(EMGA)开发了一种快速定量筛查即食海参样品中活性副溶血弧菌(V.parahaemo-lyticus)的方法.取1mL即食海参固形物伴液(在袋装的即食海参商品中,煮海参的汤即为固形物伴液),加入预装有同体积2×TCBS选择性培养基的无菌检测管,通过EMGA测定细菌生长动力学曲线和达到最大生长速率所需时长(Tmgr).以质控菌株ATCC 17802 为例,在101~105 CFU/mL范围内,活细菌浓度的对数值(log C)与Tmgr呈良好线性关系,工作曲线为Tmgr(min)=-48.822 log C(CFU/mL)+443.05(r=0.999 2),检测限为101 CFU/mL.当V.parahaemo-lyticus浓度为5.0×104 CFU/mL时,回收率为89.37%~96.06%,总检测时间<4h.该方法仅需1 种便携式仪器和3 个简便的操作步骤,为实现现场定量筛查即食海参样品的活性弧菌污染提供了可选的解决方案.[中国渔业质量与标准,2025,15(1):10-18]。
In this study,based on an electronic microbial growth analyzer,a simple and rapid method for the quantita-tive screening of viable Vibrio parahaemolyticus.contamination in ready-to-eat sea cucumber products was devel-oped.The liquid companion was sampled from the package of ready-to-eat sea cucumber product,and was directly transferred into a test tube,in which TCBS broth was preloaded.The test tube was then positioned in the electronic microbial growth analyzer to determine the bacterial growth curve,growth rate curve and the time required to reach the maximum growth rate(Tmgr).Using the QC strain as a model Vibrio parahaemolyticus,there was linear relationship between the logarithmic value of viable bacterial concentration(log C)and Tmgr over the range of 5×101~5×105 CFU/mL,with a detection limit of 101 CFU/mL.The regression equation was Tmgr(min)=-48.822 log C(CFU/mL)+443.05(r=0.999 2).When Vibrio parahaemolyticus contamination reached 5.0×104 CFU/mL,the recovery rates were 89.37%to 96.06%.The total time consumption,from the unwrapping of the package of ready-to-eat sea cucumber product to the readout of bacterial concentration,was no more than 4 h.This new method re-quires only a portable instrument and three simple manual steps,making it suitable for on-site analysis.The pro-posed method represents a promising alternative for the on-site screening and quantification of viable Vibrio spp.in ready-to-eat sea cucumber products.[Chinese Fishery Quality and Standards,2025,15(1):10-18].
