褪黑素通过抑制铁死亡改善环磷酰胺和白消安诱导的早发性卵巢功能不全小鼠的研究OA
Research on melatonin improving cyclophosphamide and busulfan-induced premature ovarian insufficiency in mice by inhibiting ferroptosis
目的 探讨褪黑素通过抑制铁死亡途径对环磷酰胺和白消安诱导的早发性卵巢功能不全(POI)小鼠的保护作用及机制.方法 选用6周龄CD1雌性小鼠,随机分为对照组、POI组和POI+褪黑素组.建立POI模型后,检测小鼠体重、卵巢湿重、血清激素水平、卵泡计数及铁死亡相关指标.结果 POI组小鼠体重、卵泡数量及激素水平显著低于对照组(P<0.05),铁死亡标志物丙二醛、核因子E2相关因子2、溶质载体家族7成员11、谷胱甘肽过氧化物酶4均升高(P<0.01).褪黑素干预后,小鼠体重、卵泡数量、血清激素水平均显著改善(P<0.05),铁死亡相关标志物丙二醛、谷胱甘肽及核因子E2相关因子2表达水平显著降低(P<0.01).结论 褪黑素通过抑制铁死亡途径改善POI,具有潜在临床应用价值.
Objective To investigate the protective effect and mechanism of melatonin on cyclophosphamide and busulfan-induced premature ovarian insufficiency(POI)in mice by inhibiting ferroptosis pathway.Methods 6-week-old CD1 female mice were randomly divided into the control group,the POI group,and the POI+melatonin group.After establishing the POI model,mouse body weight,ovarian wet weight,serum hormone levels,follicle count,and ferroptosis related indices were detected.Results The weight,follicle count,and hormone levels of the POI group mice were significantly lower than those of the control group(P<0.05),and the ferroptosis markers malondialdehyde,nuclear transcription factor E2 related factor,recombinant solute carrier family 7,member 11,and glutathione peroxidase 4 all increased(P<0.01).After melatonin intervention,the body weight,follicle count,and serum hormone levels of mice all significantly improved(P<0.05),while the expression levels of ferroptosis related markers malondialdehyde,glutathione,and nuclear transcription factor E2 related factor were significantly reduced(P<0.01).Conclusion Melatonin improves POI by inhibiting the ferroptosis pathway and has potential clinical application value.
