人参固本口服液对佐剂关节炎大鼠的抗炎作用研究OA
Study on the anti-inflammatory effect of Renshen Guben Oral Liquid on rats with adjuvant arthritis
目的 探讨人参固本口服液对佐剂关节炎(AA)大鼠的抗炎及保护作用机制.方法 建立AA大鼠模型,分别设空白组(K组)、甲氨蝶呤(J组)、低浓度人参固本口服液组(R1 组)、高浓度人参固本口服液组(R2组)、人参固本口服液+甲氨蝶呤联合组(R3 组),每组各 8 只,分别给药干预 21 d.在造模时和第 7、14、21、28 天测定大鼠左足踝关节周径并进行关节炎指数评分,ELISA法检测血清中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-17(IL-17)、血管内皮生长因子(VEGF)及肿瘤坏死因子-α(TNF-α)水平.结果 治疗 21 d后,J组、R2 组及R3 组AA大鼠关节炎指数评分及左足踝关节周径低于M组,差异有统计学意义(P<0.05).J组及R3组IL-1β、IL-6、IL-17、VEGF水平低于M组,差异有统计学意义(P<0.05);R2 组IL-1β、IL-6 水平低于M组,差异有统计学意义(P<0.05);R1 组IL-1β水平低于M组,差异有统计学意义(P<0.05).结论 人参固本口服液对AA大鼠有一定的治疗保护效果,可能与降低IL-1β、IL-6、IL-17、VEGF及TNF-α等炎性因子水平有关,且与剂量及联用甲氨蝶呤密切相关,两者联用在AA大鼠关节保护及抗炎调节作用上可能具有互补的药理活性.
Objective To investigate the mechanism of anti-inflammatory and protective effects of Renshen Guben Oral Liquid on rats with adjuvant arthritis(AA).Methods An AA rat model was established with the blank group(Group K),the methotrexate group(Group J),the Renshen Guben Oral Liquid low-concentration group(Group R1),the Renshen Guben Oral Liquid high-concentration group(Group R2),and the Renshen Guben Oral Liquid+methotrexate combination group(Group R3),respectively,with 8 rats in each group.These rats were given the drug intervention for 21 days,respectively.At the time of modeling and on Days 7,14,21 and 28,the circumference of the left ankle joint of the rats was measured and scored by arthritis index.The serum levels of interleukin-1β(IL-1β),interleuki-6(IL-6),interleukin-17(IL-17),vascular endothelial growth factor(VEGF),and tumor necrosis factor α(TNF-α)were detected by ELISA.Results After 21 days of treatment,the arthritis index score and the circumference of the left ankle joint of AA rats were lower in groups J,R2 and R3 than those in group M,with statistically significant differences(P<0.05).The levels of IL-1β,IL-6,IL-17 and VEGF were lower in groups J and R3 than those in group M,with statistically significant differences(P<0.05).The levels of IL-1β and IL-6 were lower in group R2 than those in group M,with statistically significant differences(P<0.05).The level of IL-1β was lower in group R1 than that in group M,with a statistically significant difference(P<0.05).Conclusion Renshen Guben Oral Liquid has certain therapeutic and protective effects on AA rats,which may be related to the reduction of the levels of inflammatory factors,such as IL-1β,IL-6,IL-17,VEGF and TNF-α,and is closely related to the dose and the co-administration of methotrexate.Co-administration of the two drugs may have complementary pharmacological activities on the joint protection and anti-inflammatory regulation effects in AA rats.
