有机肥和海藻酸配施提高盐碱地蒙古黄芪抗逆性和品质OA北大核心CSTPCD

Combined application of organic fertilizer and alginic acid improves the stress resistance and quality of Astragalus mongolicus in saline-alkali land

[目的]研究有机肥施用量和海藻酸施用浓度对盐碱地蒙古黄芪[Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao]光合效率、抗逆能力、产量和质量的影响,为盐碱地中蒙古黄芪生态种植的合理施肥提供理论参考.[方法]以蒙古黄芪种苗为材料,连续两年在内蒙古固阳县的盐碱地上开展了大田试验.试验采用两因素三水平完全设计,两因素为有机肥(O)、海藻酸(A).3 个有机肥施用量为 0、1200、2400 kg/hm2,分别记为O0、O1、O2;3 个海藻酸施用浓度为 0、1、2 g/L,分别记为A0、A1、A2,共组成 9 个处理.于蒙古黄芪种苗返青后 60 天开始测定叶片光合效率参数、抗氧化酶活性和渗透调节物质含量,于蒙古黄芪种苗返青后 120 天开始测定根系产量、药效成分和重金属含量.[结果]相较于O0 处理,O1、O2 处理黄芪两年净光合速率(Pn)、蒸腾速率(Tr)和气孔导度(Gs)分别提高了 13.6%~29.2%、17.0%~32.3%、14.8%~21.2%,过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、过氧化物酶(POD)活性分别提高了 10.4%~24.0%、9.5%~13.5%、7.5%~29.5%,可溶性糖(SS)、可溶性蛋白(SP)、脯氨酸(Pro)含量分别提高了 11.0%~28.8%、22.3%~94.3%、13.2%~61.4%,根系产量提高了 3.8%~8.1%,黄芪甲苷(AIV)和毛蕊异黄酮葡萄糖苷(C7G)含量分别提高了 7.4%~19.4%、4.5%~22.7%,Pb、Cd、As和Cu含量分别下降了 1.4%~16.0%、5.0%~19.5%、5.6%~9.7%、11.5%~35.2%.相较于A0 处理,A1 处理显著提高了黄芪光合效率、抗氧化酶活性和渗透调节物质含量,提升了根系产量和药效成分含量,Pb、Cd、As、Hg 和 Cu 含量分别降低了 12.0%~22.2%、6.9%~17.1%、7.5%~13.3%、7.1%~18.9%、16.1%~21.9%.2022、2023 年均以O2A1 处理的产量(6887、7153 kg/hm2)最高,O0A2处理的产量(6065、6278 kg/hm2)最低.2022、2023年O1A1处理的两种药效成分含量最高,AIV含量(0.144%、0.148%)较O2A1 提高了 5.1%~7.2%,C7G含量(0.059%、0.060%)较O2A1 处理提高了 5.3%~13.5%.[结论]施用有机肥和适宜浓度的海藻酸均可增强蒙古黄芪的光合效率,提高蒙古黄芪的抗逆能力,进而提高蒙古黄芪的产量和药用品质.以施用有机肥 1200 kg/hm2 配合喷施浓度为 1 g/L的海藻酸提升盐碱地蒙古黄芪产量、品质和抗逆能力的效果最佳,且没有引起重金属污染的风险.

[Objectives]Organic fertilizer and alginic acid have been proved of increasing the stress resistance of crop.We studied the combinations of organic fertilizer rates and alginic acid concentrations on improving the photosynthetic efficiency,stress resistance,yield,and medical quality of A.mongholicus in saline-alkali land.[Methods]A field experiment with a two-factor-three-level complete design was conducted in saline-alkali land for two consecutive years,using seedlings of A.mongholicus as materials.The two factors were organic fertilizer and alginic acid,the three application levels of organic fertilizer were 0,1200,and 2400 kg/hm2(denoted as O0,O1,and O2);and the three application concentration levels of alginic acid(A)were 0,1,and 2 g/L(denoted as A0,A1,and A2).At 60 days after the rejuvenation of A.mongholicus,the leaf photosynthetic parameters,antioxidant enzyme activities,and osmoregulation substance contents were measured.And at 120 days,the yield,and the contentd of medicinal components and heavy metals in A.mongholicus roots were measured.[Results]Compared with O0,O1 and O2 treatment increased the leaf Pn,Tr,and Gs in 2022 and 2023 by 13.6%-29.2%,17.0%-32.3%,and 14.8%-21.2%,enhanced the activities of CAT,SOD,and POD by 10.4%-24.0%,9.5%-13.5%,and 7.5%-29.5%,increased of soluble sugar,soluble protein,and proline contents by 11.0%-28.8%,22.3%-94.3%,and 13.2%-61.4%,and increased the root yields by 3.8%-8.1%,the AIV contents by 7.4%-19.4%and C7G contents by 4.5%-22.7%,and decreased Pb,Cd,As,and Cu contents by 1.4%-16.0%,5.0%-19.5%,5.6%-9.7%,and 11.5%-35.2%,respectively(P<0.05).Compared with A0,A1 treatment significantly increased all the tested photosynthesis parameter values,the activities of antioxidant enzymes,and the contents of osmoregulation substances,significantly enhanced the root yields and medicinal component contents,and decreased Pb,Cd,As,Hg,and Cu contents by 12.0%-22.2%,6.9%-17.1%,7.5%-13.3%,7.1%-18.9%,and 16.1%-21.9%,respectively.In both 2022 and 2023,O2A1 treatment was recorded the highest root yield(6887,and 7153 kg/hm2)while O0A2 treatment was recorded the lowest root yield(6065,and 6278 kg/hm2).Compared with O2A1 treatment,O1A1 increased AIV content by 5.1%-7.2%and C7G content by 5.3%-13.5%.[Conclusions]Both organic fertilizer and alginic acid application showed obvious effect on increasing the stress resistance,root yield and the medicinal composition contents of A.mongholicus,due to the enhanced photosynthetic efficiencies and antioxidant enzyme activities.Satisfactory synergetic effect on yield and quality but low heavy metal content can be obtained at combination rate of organic fertilizer 1200 kg/hm2 with 1.0 g/L of alginic acid to produce A.mongholicusin saline-alkali land.
