响应面法优化提取壮药三七姜总酚酸及其镇痛抗炎作用机制研究OA北大核心CSTPCD

Optimization of response surface method for the extraction of Zhuang medicine Stahlianthus involucratus total phenolic acids and its analgesic and anti-inflammatory mechanism

试验采用响应面法优化壮药三七姜总酚酸提取工艺并探究其镇痛抗炎作用机制.以三七姜为原料,采用回流提取法提取总酚酸,通过单因素试验和Box-Behnken试验对提取工艺进行优化,探究乙醇浓度、液料比、提取时间对总酚酸得率的影响;建立醋酸致小鼠扭体模型和小鼠棉球肉芽肿模型,探究三七姜总酚酸的镇痛抗炎作用机制.结果表明,三七姜总酚酸的最优提取工艺为乙醇浓度42%、液料比60mL/g、回流时间60 min.在此条件下,三七姜总酚酸得率可达6.09 mg/g.与模型组相比,三七姜醇提物各剂量对肉芽肿炎症具有抑制作用,可以降低小鼠扭体次数,降低白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、前列腺素E2(PGE2)、白细胞介素-1β(IL-1β)、5-羟色胺(5-HT)含量,增加环磷酸腺苷(cAMP)含量.研究表明,响应面法优化壮药三七姜总酚酸的提取工艺稳定可靠;三七姜总酚酸具有一定的镇痛抗炎活性,其机制可能与减少体内IL-6、TNF-α、IL-1β和PGE2等促炎性细胞因子分泌有关.

Response surface method(RSM)was used to optimize the extraction process of Stahlianthus involucratus total phenolic acids and explore its analgesic and anti-inflammatory mechanism.Total phenolic acids were extracted from Stahlianthus involucratus by reflux extraction.The extraction process was optimized by single factor test and Box-Behnken test,and the effects of ethanol concentration,liquid-to-material ratio,and extraction time on the yield of total phenolic acids were explored.Writhing model and cotton ball granuloma model of mice induced by acetic acid were established to study the analgesic and anti-inflammatory mechanism of total phenolic acids in Stahlianthus involucratus.The results showed that the optimum extraction process of total phenolic acids from Stahlianthus involucratus were an ethanol concentration of 42%,liquid-material ratio of 60 mL/g,and reflux time of 60 min.Under these conditions,the yield of total phenolic acids of Stahlianthus involucratus can reach 6.09 mg/g.Compared with the model group,each dose of ethanol extract of Stahlianthus involucratus had an inhibitory effect on granuloma inflammation,reduced the number of writhing,decreased the contents of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),prostaglandin E2(PGE2),interleukin-1β(IL-1β),5-hydroxytryptamine(5-HT)and increased the content of cyclic adenosine monophosphate(cAMP).The study indicates that the optimized extraction process of Stahlianthus involucratus by response surface methodology is stable and reliable,and can be used for the extraction of total phenolic acids from Stahlianthus involucratus.Stahlianthus involucratus has certain analgesic and anti-inflammatory activities,and its mechanism may be related to reducing the secrete of pro-inflammatory cytokines such as IL-6,TNF-α,IL-1β,and PGE2 in vivo.
