荆鲜止痒涂剂治疗犬皮肤瘙痒症的作用机制探讨OA

The mechanism of Jingxian Zhiyang Tuji in treating canis skin pruritus

采用网络药理学方法和分子对接技术探讨荆鲜止痒涂剂治疗犬皮肤瘙痒症的作用机制.通过TCMSP筛选有效成分及其作用靶点,将靶点导入Uniport数据库构建药物-成分-靶点网络,从GeneCards和OMIM数据库查找与犬皮肤疾病、皮肤瘙痒相关的基因,与药物活性成分的作用靶点建立交集后,采用STRING数据库和Cytoscape3.9软件绘制蛋白相互作用网络并构建PPI网络图,可视化后筛选核心靶点,利用DAVID数据库对药物、疾病共同靶点进行GO功能注释和KEGG富集分析.最后通过分子对接技术对预测出的活性成分和核心靶点进行分子对接验证.结果筛选得到146个活性成分,主要成分为槲皮素、木犀草素、芹菜素、芒柄花黄素、汉黄芩素、β-谷甾醇、西托糖苷、槐香碱、金丝桃素、异橙黄酮.与犬皮肤瘙痒症相关的靶点有55个,其中核心靶点为TNF、IL-6、VEGFA等.核心靶点参与细胞因子活性、细胞外隙、细胞凋亡、RNA聚合酶Ⅱ启动子转录的正调控等生物过程的调节,作用于调控MAPK信号通路、细胞因子-细胞因子受体相互作用信号通路、IL-17信号通路等,通过抗炎、抑菌、抗氧化等过程减轻犬皮肤瘙痒的症状.分子对接结果显示药物的关键成分与核心靶点有良好的结合性.表明荆鲜止痒涂剂对犬皮肤瘙痒症的治疗涉及多成分、多靶点、多通路的机制.

In order to investigate the mechanism of action of Jingxian Zhiyang Tuji in the treatment of canis skin pruritus,network pharmacology method was adopted in this experiment.The active ingredients and their targets were screened by TCMSP,and all the targets were imported into Uniport database to form a drug-composition-target network.Genes related to canis skin diseases and pruritus were searched from GeneCards and OMIM databases.After intersection with the target of active drug ingredients,protein-protein interaction was mapped using STRING database and Cytoscape3.9 software.PPI network map was constructed and core targets were screened after visualization.Finally,GO and KEGG enrichment analysis was performed on common targets of drugs and diseases using DAVID database.The predicted active ingredients and core targets were verified by molecular docking technology.The results showed that 146 active ingredients were obtained,and the main components were quercetin,luteolin,apigenin,baicalein,β-sitosterol,cetoposide,sophorine,hypericin and isohesperetin.There were 55 targets associated with canine pruritus,among which the core targets were TNF,IL-6,VEGFA,etc.The core target is involved in the regulation of biological processes such as cytokine activity,extracellular space,apoptosis process,and positive regulation of RNA polymeraseⅡpromoter transcription,and acts on the regulation of MAPK signaling pathway,cytokine-cytokine receptor interaction signaling pathway,IL-17 signaling pathway and other signaling pathways in the anti-inflammatory,antibacterial,antioxidant and other processes to alleviate canis skin pruritus symptoms.The results of molecular docking showed that the key components of the drug had good binding with the core target.It showed that the treatment of canis pruritus with Jingxian Zhiyang Tuji paint involves multi-component,multi-target and multi-pathway mechanisms.
